Item Effects of glucosinolates and their hydrolysis products on energy balances and performance parameters and histological parameters in broiler chicken diets(Bursa Uludağ Üniversitesi, 2018-07-17) Belenli, Deniz; Berhow, Mark A.; Polat, Umit; Erdost, Hatice; İlhan, Tuncay; Orman, Abdülkadir; Yesilbağ, Derya; Veteriner Fakültesi; Histoloji ve Embriyoloji Ana Bilim DalıGlucosinolates are important bioactive molecules and widely found in Brassicaceae species (cress, brussels sprouts, mustard, broccoli, kale etc.). Depending on the amount of these vegetables consumed, both positive and negative metabolic effects from glucosinolate metabolites may occur. The aims of this study were to; investigate inexpensive animal food sources that both increases weight gain and provides enhanced performance parameters without adversely affecting the animal’s health and metabolism; to evaluate dose adjustment of food containing glucosinolates in animals; and to evaluate changes in the biochemical and performance status of chickens on these glucosinolate containing diets. A total of 624 one-day-old Ross 308 broiler line chicks were divided into one control and three treatment groups. Cress seed (Lepidium sativum) was added 0.05% for the first treatment groups (Group 1, 10 g/kg), 0.10% for the second treatment groups (Group 2, 20 g/kg) and 0.15% for the last treatment groups (Group 3, 30 g/kg) to the diet. Serum samples were evaluated for serum glucose, adiponectin, leptin, growth hormone, estradiol and cortisol levels. Performance parameters investigated included feed intake, live body weight and feed conversion ratio. . The villus length, number of goblet cells, crypt depth were determined for histological analyses. According to histological results, villus length was significant at p < 0.05 level between control and group 1 and at p < 0.001 level with groups 2 and 3 at 21 days. The depth of the crypts belonging to the control and experimental groups was not significant between the control group and the group 1 when the statistic was evaluated on the 21st day, whereas between the control group and the group 2, p < 0.05 and p < 0.01; statistical significance was found at p < 0.001 level between group 3 and control group. The results showed that dietary glucosinolate supplementation as feed additive (10, 20 and 30 g/kg) did not significantly improve the dietary performance, or carcass parameters of broiler chickens. Feed intake was the highest in group 2 (20 g/kg), female live weight was the highest in group 2 (20 g/kg) and 3 (30 g/kg). In conclusion, the rates of the cress seed (0.05, 0.10 and 0.15%) that contain glucotropaeolin were not affected for feed additive on performance (especially live weight and live weight gain) and carcass parameters.Item Serum biochemical profile of broiler chickens fed diets containing rosemary and rosemary volatile oil(Uludağ Üniversitesi, 2011) Polat, Ümit; Yeşilbağ, Derya; Eren, Mustafa; Veteriner Fakültesi; Biyokimya Ana Bilim DalıThe study was conducted to determine the effects of dietary supplementation rosemary aromatic plant, rosemary volatile oil and α-tocopherol acetate (Vitamin E) on serum variables of broilers fed on maize-soybean meal based diets. Eight hundred 1-d-old Ross-308 male chickens were weighed and randomly divided into 1 control and 7 experimental groups each with 10 replicates of 10 birds. There were 8 dietary treatments: (VitE1) control without rosemary and rosemary volatile oil only with 50 mg/kg vitamin E; (R1) 5.7 g/kg ground rosemary leaves; (R2) 8.6 g/kg ground rosemary leaves; (R3) 11.5 g/kg ground rosemary leaves; (RO1) 100 mg/kg rosemary volatile oil; (RO2) 150 mg/kg rosemary volatile oil; (RO3) 200 mg/kg rosemary volatile oil and (VitE2) 200 mg/kg vitamin E. Broilers consumed the diets and water ad libitum. After 42 days, 80 animals were randomly selected for serum biochemical profile analysis involving ceruloplasmin, superoxide dismutase activity (SOD), transferring, albumin globulins ratio (A/G), total cholesterol, creatin, urea, alanine aminotransferase (ALT) and aspartate amino transferase (AST). While serum transferrin, urea level and ALT-AST activity were not statistically different among groups serum ceruloplasmin (p< 0.000), SOD activity (p<0.05), albumin/globulin ratio (p< 0.000), total cholesterol (p<0.001), creatinin (p<0.05) and AST (p< 0.000) level were found to be significantly different. In conclusion, the Rosmarinus officinalis plant and its volatile oil have increasing effect on serum SOD activity and effect positively oxidation mechanism. On the other hand, it can be assumed that rosemary plant created hypocholesterolemic effect in this study.Item Serum tumor necrosis factor - alpha alterations of dry cows vaccinated with inactive coronavirus, rotavirus, E. coli combined vaccine(Uludağ Üniversitesi, 2017-04-05) Mecitoğlu, Zafer; Akgül, Gülşah Demir; Catik, Serkan; Udum, Duygu; Batmaz, Hasan; Veteriner Fakültesi; İç Hastalıkları Ana Bilim DalıTumor Necrosis Factor alpha (TNF-α) is a proinflamatory cytokine, closely associated with insulin resistance and related diseases such as type II ketosis and fatty liver of cows. Aim of the presented study was to investigate circulating levels of TNF-α after vaccinating dry cattle with inactive rotavirus, coronavirus and E.coli combined vaccine. 14 cattle were randomly assigned to two groups; vaccination group was vaccinated on days 0 and 21 of the study and control group not. Blood samples were collected on days 0, 1, 5, 15, 21, 22, 26, 36 of the study for evaluation of TNF-α and total leukocyte counts. Although not significant, TNF-α levels decreased in vaccination group after the second application. Total Leukocyte counts did not differ between and within groups. Results of the study demonstrates that vaccinating cattle with inactive rotavirus, coronavirus and E.coli combined vaccine in dry period does not induce alterations in circulating TNF-α levels.Item Trace element concentrations in breast milk and sera: Relations with lactation(Uludağ Üniversitesi, 2007-12-01) Özkan, Tanju Başarır; Durmaz, Neslihan; Erdemir, Gülin; İlçöl, Yeşim Özarda; Tıp Fakültesi; Biyokimya Ana Bilim DalıBreast milk has unique properties; one is the variability of the compounds in ratio according to the baby’s needs for every lactation period. Iron and zinc are essential elements for life. The aim of this study is to determine Fe and Zn levels in plasma and breast milk of mothers through the first 4 months after delivery and to evaluate its relation with infants’ growth and immune system functions. The study conducted through January 2000 – April 2001 in Bursa with 27 mother-infant couples. The babies grouped into two: Group 1 (n: 16) was formed by the breast milk fed babies, and Group 2 (n: 11) included the babies fed with both breast milk and formula. Besides clinical assessment of the babies, laboratory evaluations were performed on the sera of couples and breast milk. While the comparison of Zn level in colostrum with mature and late mature breast milk demonstrated a statistical significance (p<0.05), Fe had a similar trend without significance. Fe deficiency anemia was found in high percentages in both groups; 32.5% of Group1 and 27.7% of Group2 (p>0.05). Our study affirms the suggestion for Fe supplementation for healthy, term infants beginning from 4th month regardless of nutritional model.
