Abstract
Bu tez çalışmasında, bakır(II) iyonunun amino asit Schiff bazı ligandı içeren komplekslerini sentezlemek için, aminoasitlerden L-triptofan, salisilaldehit türevlerinden5-klorosalisilaldehit ve 3,5-diklorosalisilaldehit ve ayrıca nötral ligant olarak 1,10-fenantrolin seçildi. Tezin ilk aşamasında; bakır(II) iyonunun triptofan Schiff bazları ve 1,10-fenantroliniçeren ikili ve karışık ligant kompleksleri sentezlendi. L-triptofan ve 5-klorosalisilaldehitten/3,5-diklorosalisilaldehitten türeyen tripto fan Schiff bazı ligandları; kompleks sentezi sırasında çözelti ortamında oluşturuldu. Sentezlenen komplekslerin yapıları ve özellikleri; elementel analiz, IR spektroskopisi, tek kristal X-ışını kırınımı, elektronik absorpsiyon spektroskopisi ve termik analiz yöntemleri ile belirlendi. Tezin ikinci aşamasında ise; sentezlenen komplekslerin biyolojik aktivite çalışmaları yapıldı. Komplekslerin buzağı timüsü DNA’sı olan CT-DNA ile etkileşimleri termalde natürasyon, elektronik absorpsiyon ve floresans spektroskopisi yöntemleri ile araştırıldı. Komplekslerin CT-DNA’ya bağlanma sabitleri hesaplandı. Komplekslerin DPPH radikal süpürme ve H2O2 radikali yakalama aktiviteleri standart olarak kullanılan askorbik asit, trolox ve BHT ile karşılaştırmalı olarak araştırıldı. Komplekslerin antikanser aktiviteleri ise; MCF-7 ve MDA-MB-231 insan meme kanser hücre hatlarında kemoterapik ilaç olan cis-Platin ve Paklitaksel ile karşılaştırmalı olarak belirlendi.
In this thesis, to synthesize copper(II) ion complexes containing amino acid Schiff base ligand, amino acids L-tryptophan, salicylaldehyde derivatives 5-chlorosalicylaldehyde/3,5-dichlorosalicylaldehyde and 1,10-phenanthroline as neutral ligand were selected. In the first part of the thesis; binary and mixed ligand complexes of copper (II) ion containing tryptophan Schiff bases and 1,10-phenanthroline were synthesized. tryptophan Schiff base ligands derived from L-tryptophan and 5-chlorosalicylaldehyde / 3,5-dichlorosalicylaldehyde; formed in the solution during complex synthesis. Structures and properties of synthesized complexes; determined by elemental analysis, IR spectroscopy, single crystal X-ray diffraction, electronic absorption spectroscopy and thermal analysis methods.In the second part of the thesis; biological activity studies of the synthesized complexes were carried out. The interactions of the complexes with calf thymus DNA (CT-DNA) were investigated by thermal denaturation, electronic absorption and fluorescence spectroscopy methods. The binding constants of the complexes to CT-DNA were calculated. DPPH radical scavenging and H2O2 radical scavenging activities of the complexes were investigated in comparison with standard ascorbic acid, trolox and BHT. The anticancer activities of the complexes were determined in comparison with cis-Platinum and Paclitaxel, which are chemotherapeutic drugs in MCF-7 and MDA-MB-231 human breast cancer cell lines.
