Abstract
Bu çalışma, anter kültüründe farklı tiplerde 8 farklı acı biber genotipinin 2 farklı besi ortamında haploid bitki oluşturma etkinliklerini belirlemek amacıyla yürütülmüştür. Denemede bir adet California Wonder, iki adet Anaheim, iki adet Ancho ve üç adet Jalapone acı biber genotipi kullanılmıştır. Anterler; 4 mg/L Naphthaleneacetic acid (NAA), 0,1 mg/L Benzylaminopurine (BAP), 15 mg/L gümüş nitrat (AgNO₃), 2,5 g/Laktif kömür, 30 g/L sakkaroz ve 8 g/L agar içeren Murashige ve Skoog (MS) besi ortamı(BO1) ile 4 mg/L NAA, 0,1 mg/L BAP, 15 mg/L AgNO₃, 0,5 mg/L Vitamin C, 0,05 mg/Lbiotin, 2,5 g/L aktif kömür, 30 g/L maltoz ve 8 g/L agar içeren MS besi ortamında (BO2)kültüre alınmışlardır. Çalışmada ele alınan tüm parametreler üzerinde besi ortamı vegenotip özelliklerinin etkisi istatistiksel olarak p<0.01 düzeyinde önemli bulunmuştur. Genel olarak embriyo miktarı ve bitkiye dönüşen embriyo miktarı üzerinde BO2 besi ortamından daha yüksek sonuçlar elde edilirken incelenen diğer parametrelerde BO1 besi ortamı daha yüksek sonuçlar vermiştir. Elde edilen embriyo miktarı, bitkiye dönüşen embriyo, gelişen bitki, haploid bitki ve spontane double haploid bitki miktarı genotiplere göre istatistiksel olarak %1 düzeyinde önemli farklılık göstermiştir. İncelenen tüm parametrelerde Anaheim G2 genotipi daha iyi sonuç vermiştir. Anterlerden elde edilen embriyolar belirli oranlarda bitkiye dönüşürken Ancho G4 ve Jalapeno G6 genotiplerdeki bitkiler gelişme gösterememiştir. Bu sonuçlar tek bir standart anter kültürü protokolünün farklı biber tiplerinde aynı etkinliği meydana getirmediğini, protokollerin tip/genotip bazında özelleştirilmesi gerekliliğini de ortaya koymaktadır.
This study was conducted in order to determine the activities of forming haploid plants in 2 different nutrient mediums of 8 different types of chili pepper genotypes in anther culture. One California Wonder, two Anaheim, two Ancho and three Jalapone hot pepper genotypes were used in the experiment. Anthers; 4 mg/L Naphthaleneacetic acid (NAA), 0.1 mg/L Benzylaminopurine (BAP), 15 mg/L silver nitrate (AgNO₃), 2.5 g/L activated charcoal, 30 g/L sucrose and 8 g/L agar containing Murashige and Skoog (MS) nutrient medium (BO1) with 4 mg/L NAA, 0.1 mg/L BAP, 15 mg/L AgNO₃, 0.5 mg/L Vitamin C, 0.05 mg/L biotin, 2.5 g/L activated charcoal, 30 g/L maltose and 8 g/L agar containing MS nutrient medium (BO2) were cultured. The effect of nutrient medium and genotype characteristics on all parameters considered in the study was found to be statistically significant at the p<0.01 level. In general, higher results were obtained from the BO2 nutrient medium on the amount of embryos and the amount of embryos that turned into plants, while the BO1 nutrient medium yielded higher results in other parameters studied. The amount of embryos obtained, the embryo that turned into a plant, the developing plant, the haploid plant and the amount of spontaneous double haploid plants showed statistically significant differences at the level of 1% according to genotypes. The Anaheim G2 genotype performed better in all parameters examined. While embryos obtained from anthers turned into plants at certain rates, plants of Ancho G4 and Jalapeno G6 genotypes did not develop. These results reveal that a single standard anther culture protocol does not produce the same effectiveness in different pepper types, and it is necessary to customize the protocols on the basis of type/genotype.
