Broiler damızlık, ticari broiler ve yumurta tipi tavuk işletmelerinde salmonella türlerinin izolasyon ve identifikasyonunda kullanılan metodların karşılaştırılması

Abstract

Bu çalışma. Bursa, İstanbul ve İzmir yörelerinde 20 adet damızlık, 17 adet yumurtacı ve 13 adet broiler işletmesinde salmonella izolasyonu için, kloakal swab ve Drag swab yöntemlerinin karşılaştırılması amacıyla yapıldı. Her damızlık işletmesinden 53, yumurtacılardan 100 ve broilerlerden 100 adet kloakal swab alındı. Kloakal swab tekniğinde, swab, nutrient buyyon, tetrathionate broth ve ağar kullanılırken; Drag swab metodunda özel Drag swab, Double Strength Skim Milk, XLT ağar ve BGN ağardan yararlanıldı. İzole edilen Salmonella suçları biyokimyasal ve serolojik olarak identifiye edildi. Damızlık işletmelerden kloakal swab yöntemiyle 2 CX 0.18) S. gallinarum ve 2 (X 0.18) . typhimurium izolasyonu gerçekleştirildi. Diğer işletmelerden kloakal swab yöntemiyle izolasyon sayıları şu şekildeydi: Ticari yumurtacılardan; 2 (X 0.1) S. gallinarum, 1 C/. 0.05) S. typhimurium ve 2 <7. 0.1) S. enteritidis, ticari broilerlerden ise; 1 (7. 0.7) . gallinarum, 1 C/. 0.7) . typhimurium ve 1 (X 0.7) . enteritidis izolasyonu yapıldı. Drag swab ile bu işletmelerden izolasyon ise aşağıdaki şekilde sonuçlandı. Broiler damızlıklardan; 2 (X 10.0) S. gallinarum, 3 (X 15.0) typhimurium ve 1 (X 5.0) . enteritidis, ticari yumurtacılardan; 2CX11.7) S. gallinarum, 2 (% 11.7) . typhimurium ve 3 (54 17.6). enteritidis, ticari broilerlerdenş 1 (7. 7.6). gallinarum 2 (.7. 15.3) . typhimurium ve 2 (5415.3) . enteritidis izolasyonu gerçeklestirildi. Kloakal swabla 5 S. gallinarum, 4 . typhimurium ve 3 enteritidis izolasyonu yapılırken, Drag swabla 5. gallinarum, 7 . typhimurium ve 6 .enteri t idis izole edildi. Bu çalışmanın sonucunda konakçıya spesifik olmayan salmonella izolasyonunda Drag swab tekniğinin, kloakal swab yöntemine göre daha etkin ve pratik olduğu tespit edildi. Ayrıca sağlıklı görülen sürülerde predominant salmonella serotierinin . typhimurium ve enteritidis olduğu da açıklık kazandı.Comparison of the methods using isolation and Identification of Salmonella strains in the farms of broiler parents, commercial broiler and laying hens. This work was carried out for a comparison of the procedures of Cloacal swab and Drag Swab for Salmonella isolation in 20 breseler, 17 layer and 13 broiler flocks in Bursa, İstanbul and İzmir districts. Fiftythree, 100 and 100 Cloacal Swab were taken from each of the breeder, layer and broiler flocks, respectively. Sterile Swabs, nutrient broth, tetrathionate broth and SS ağar were used in the Cloacal Swab procedure while specially designed drag swab, Double Strength Skim Milk, XLT ağar and BGN ağar were ıncluded in the Drag 4 Swab procedure. The isolated and suspected Salmonella Strains were identified biochemical ly and serologically. 2 (0.18 %) gallinarum and 2 (0.18 X) . typhimurium vere isolated from breeder flocks by the Cloacal Swab procedure. isolation numbers of Salmonellae from the other two different types of flocks vere as followss Commercial layerş 2 (0.1 X) gallinarum, 1 (0.05 X) . typhimurium and 2 (0.1 X) S. enteritidis. Commercial broiler? 1 (0.7 X) S. gallinarum 1 (0.7 X) tvohimurium and 1 (0.7 X) enteritidis. On The other hand, isolastions from the flocks by the Drag Swab Procedure were concluded as followss Broiler breederş 2 (10.0 X) S. gallinarum, 3 (15.0 7.) typhimurium and 1 (5.0 X) S. enteritidis. Commercial layerj 2 (11.7 X) S. gallinarum, 2 ( 11.7 X) S. typhimurium and 3 (17.6 X) S. enteritidis Commercial broiler? 1 (7.6 X) S. gallinarum, 2 (15.3 X) S. typhimurium and 2 (15.3 X) S. enteritidis. Five gallinarum, 4 » typhimurium and 3 » enteritidis were isolated by the procedure of Cloacal Swab, but 5 . gallinarum, 7 » typhimurium and 6 . enteritidis by Drag Swab procedure. In conclusion, it was decided that Drag Swab procedure was more reliable and practical than Cloacal Swab procedure for the isolation of non-host depandent Salmonella serotypes. In addition, it was also revealed that predominant Salmonella serotypes vere . typhimurium and » enteritidis in the healthy flocks.
