Abstract
Bu araştırmada 40 mg/gün aspirinin tek doz ve uzun süre (15 gün) kullanımı sonucu tromboksan A2 (TxA2) ve prostasiklin (PGI2) in plazma düzeyleri üzerindeki etkileri araştırmaya çalışıldı. 10 sağlıklı gönüllüde uygulanan bu çalışmada, hiç ilaç kullanmadan, bir defa 40 mg/gün aspirin alındıktan sonra ve 15 gün süreyle 40 mg/gün aspirin kullandıktan sonra, toplam 3 kez kan alındı. Aspirin olarak "Phizer" finansının Babyprin 80 mg tabletlerinin yarısı verildi. Daha sonra RİA yönteminin uygulandığı NEN finansının trityum işaretli TxP 2 ve 6-keto PGF1a kitleri ile plazmada TxB2 ve 6-keto PGF1a. miktar belirtileri yapıldı, 6-keto PGF1a düzeyleri sırasıyla (X - SD), 97.5 ± 29.3, 71.7 ± 127 ve 37.75 ± 9.96 pg/ml olarak saptandı.. TxB2 def!erleri ise gene sırasıyla 88.75 ± 7.8, 39.8 12.7 ve 1.45 ± 7.6 pg/ml idi. Tek doz ve 15 günlük aspirin kullanımı sonucu TxB2'de gözlenen inhibisyon miktarları sırasıyla % 55.2 ve % 912.6 iken, 6-keto PGF2a.'da % 26.4 ve % 61.3'tü. Çalışmamızın sonucunda, atheroskleroz (As) lu ve As risk grubundaki hastalarda 40 mg/gün aspirin kullanımının, profilaktik olarak uygun olduğu kanısına varıldı.
In this study, il has been searched for the long-term (15 days) usage and single-dose of 40 mg/gün aspirins effects on plasma thromboxane A2 (TxA2) and prostacyclin (PGI2) levels. The study was applied on 10 healthy volwıteers. Their bloods are taken 3 times: Before taking any drugs, once after taking 40 mg/day and at the end of 15th day taking 40 mg/day aspirin. Half of the Babyprin 80 mg tablets of "Phizer" firm are used as aspirin. Later on, TxB and 6-keto PGF2a. amounts in plasma have been measured with the method of RIA. with the NEN firm's tritium Labeled TxB2 and 6-keto PGF2a. kits. 6-keto PGF2a. levels in plasma were respectively (X ± SD), 97.5 ± 29.3, 71.75 ± 12.7 and 37.75 ± 9.96 pg/ml. TxB2 levels were again respectively 88.75 ± 7.8, 39.8 ± 12.7 and 7.45 ± 7.6 pg/ml. The inhibition levels observed in TxB2 resulting from the usage of the single dose and 15 days term aspirin were respectively % 55.2 and % 91.6, whereas 6-keto PGF2a. were % 26.4 and % 61.3. The results of our studies have pointed out that 40 mg/day aspınn usage, can be practically appropriate, for the patients with atherosclerosis (As) and for the persons who are in the As risk group.
