| dc.description.abstract | Bu çalışmada arpa ve buğday ağırlıklı olarak hazırlanan rasyonlara yem katkı maddesi olarak enzimlerin katılmasıyla karaciğerde meydana gelebilecek değişikliklerin histolojik ve histoşimik yöntemler ile saplanmasını amaçladık. Çalışmamızda günlük-etlik civcivler yedi farklı rasyonla beslenerek, denemenin 15., 21. ve 40. günlerinde karaciğer numuneleri alınmıştır. Alman numuneler histoloji tekniğine uygun işlemlerden geçirilerek parajinde bloklanmıştır. Hazırlanan histolojik kesitlerin bir kısmına yapısal özelliklerin belirlenmesi için Crossmon'ın üçlü boyama tekniği, diğer bir kısmına ise glikozaminoglikanların belirlenebilmesi için Mc. Manus'un Periodio-Acide Schiff (PAS) metodu uygulanmıştır. Karaciğerde mevcut lipid varlığının belirlenmesi amacıyla da Sudan Black-B metodu kullanılmıştır.15 günlük yaştaki civcivlerden hazırlanan karaciğer preparatlarında normal histolojik yapı dışında herhangi bir değişiklik görülmemiştir. Mısır ile beslenen 21 günlük yaştaki grubun hepatositlerinde hücre içi vakuoller saptanmıştir. Mısır, arpa, buğday, buğday+arpa ağırlıklı enzimsiz rasyonla beslenen kırk günlük yaştaki civcivlerin hepatositlerinde vakuoliasyonun oldukça arttığı görülmüştür. Enzim katılan özellikle arpa enzimli grupta, buğday enzimli ile buğday+arpa enzimli gruplarda glikozaminoglikan birikiminde belirgin artış belirlenmiştir. Enzimli rasyon ile beslenen gruplarda karaciğer hepatositlerinin orta derecede lipid içerdiği saptanmıştır. Çalışmamızda rasyonda kullanılan enzim katkı maddelerinin karaciğerde herhangi bir dejenerasyona sebebiyet vermediği ve karaciğer yağlanmasını azalttığı sonucuna varılmıştır. | tr_TR |
| dc.description.abstract | The study was conducted to investigate the effects of enzyme supplementation of a diet based on barley and wheat on the livers of the chicken Liver samples were taken from the broiler chickens at the 15th, 21st and 40th days after feeding with 7 different diets. Rutine histolojical methods applied to the samples and then paraffine embedded. Some of the sections were stained by Crossmon's Triple Stain for structural properties and the others by Mc Manus's Periodic - Acide Schiff (PAS) for glycozaminoglicans. Sudan Black - B stain was applied to determine the lipid content of the liver. Apart from normal histological structures, no changes could be seen on the liver sections prepared from the 15th day old chicks. Some vacuols were observed in the hepatoeytes of the livers of the 21sı day - old chicks feda diet based on corn. An increasing vacuolisation was rather in the hepatocytes of the 4olh day - old chickens fed with a died based on corn , barley, wheat and barley+wheat. Glycozaminoglican deposition was increased in the chicks fed with a diet supplemented with enzyme based on barley, wheat and barley+ wheat respectively. A moderate amount of lipid was seen in the hepatocytes of the chickens fed with the enzyme supplemented diets. Any dejeneration wasn't observed in the livers of the chickens fed with enzyme supplemented diets. Enzyme supplementation to the diets decreased lipid infiltration in the liver. | en_US |
