Abstract
Endometriozis genel popülasyonun %2 ile %10'unu etkileyen, patofizyolojisi hala kesin olarak aydınlatılamamış bir hastalıktır. Endometriozis benign bir hastalık olmasına rağmen invazyon, uzak organlara lenfatik yayılım ve anjiogenez gibi malign özellikler göstermekte ve nadir de olsa malign transformasyona uğramaktadır. Endometriozisli hastalarda artmış oksidatif stres parametreleri kanıtlanmasına rağmen medikal tedavi seçenekleri kısıtlıdır. Bu deneysel çalışmada, antioksidan tedavinin(vitamin C ve E), endometriotik odak hacmi ve hücre DNA hasarı üzerine etkisinin araştırılması hedeflendi. Bu çalışmada, toplam 20 adet, ortalama 250 gram ağırlığındaki, Wistar Albino dişi sıçan kullanıldı. Kontrol Grubu: Otolog endometriozis modeli + serum fizyolojik (10 sıçan) Deney Grubu: Otolog endometriozis modeli + vitamin C ve E (10 sıçan) Her iki grupta endometriozis modeli oluşturulduktan sonra intraperitoneal enjeksiyonlara başlandı. 31 günlük çalışma süresi sonunda gruplar ağırlık, endometriotik odak hacmi, gland-stroma skoru ve DNA hasarı(TUNEL(+)lik oranı) olarak karşılaştırıldı. Toplamda 11 deney hayvanı ile çalışma tamamlandı. Analiz edilen veriler ile gruplar arası anlamlı fark bulunamadı(p>0.05). İstatistiksel olarak anlamlı sonuç elde edilemese de analize dahil edilen deney hayvanı sayısı genişletilerek farklı, anlamlı sonuçlar kazanılabilir.
Endometriosis effects 2 to 10% of the general population but its pathophysiology still remains unknown. Even though endometriosis is a benign disease, malignancy criteria such as, invasion, distant organ involvement and angiogenesis are present. In cases less than %1, malignant transformation is detected. The presence of elevated levels of oxidative stress parameters in patients with endometriosis are well known, nevertheless medical treatment options are limited. In this experimental study, we aimed to investigate the effect of antioxidant treatment(vitamin C and E) on endometriotic cyst volume and cell DNA damage. In this study, a total of 20 Wistar Albino rats, mean weights of 250 gr, were used. Control Group: Autologous endometriosis model + serum physiologic(10 rats) Study Group: Autologous endometriosis model + vitamin C and E(10 rats) Intraperitoneal injections were started after the endometriosis model was completed. At the end of 31 days of study period, weights, endometriotic cyst volume, gland-stroma score and DNA damage(TUNEL(+) percentage) were compared between the two groups. The study was finalized with 11 rats. Analized data did not show any significant difference between groups(p>0.005). Although these differences were short of statistical significance, increasing the scope of the study may alter the results.
