Abstract
Bu çalışmada, bir promutajen olan 2-aminoflorenin biotrans- formasyon hızının tayininde ve aynı maddenin mutajenik türevlerinln ölçümünde spektroskopik yöntemler yerine modifiye edilmiş Salmonella-mikrozom mutasyon testinin uygulanabilirliği araştırıldı. Yapılan deneylerde S.typhi murium LT.2 susunun TA 98 straini kullanıldı. Bu strainin genotipik özelliklerinin kontrolü ve mikrozomların hazırlanması Maron ve Ames (I9ö3)'e göre yapıldı. Daha sonra, 2-aminoflorenin bi o transformasyon hızı 3 değişik şekil de ölçüldü. Uyguladığımız deney şartlarına göre maksimum trans - formasyon hızına 25-30. dakikalarda ulaşıldığı görüldü. Elde edilen sonuçlardan sonra metabolik aktivasyon olaylarının ölçümünde, pahalı spektroskopik yöntemler yerine, bizim modifiye ettiğimiz şekliyle Salmonella-mikrozom testinin kullanılabileceği görüldü.
In this study, the determination of the rate of biotrans formation of the promutajen 2-aminoflorene and the possibili ties of the use of modified Salmonella-mi crosorae mutagenicity test for measuring biotransformed mutagenic derivatives of 2-aminoflorene, instead of spectroscopic methods were investigated. Salmonella typhimurium LT. 2 »tester strain TA 98 was used throughout the experiments. Strain checking and microsome pre paration were done according to the recommendations of Maron and Ames (1983). The rate of 2-aminoflorene biotransformation was determined by using three different methods. The results ob tained showed that under our experimental conditions the time to achive the maximum transformation rate was reached by 25 to 30 minutes. Thus it appears that, our modified Salmonella-mi crosome mu tagenicity test can be effectively substituted in place of the more elaborate and expensive spectrophotometric methods for measurements of biotransformation events.
