Abstract
Bu çalışmada, antiprotozoal bir ilaç ve folik asit antogonisti olan pyrimethamine'nin insan kromozomlarında oluşturduğu hasara karşı askorbik asit ve folinik asidin antiklastojenik etkileri araştırıldı. Deney grubu olarak, sigara ve alkol kullanmayan 18 sağlıklı kişi seçildi. Bireylerden alınan periferik kanlarla hazırlanan lenfosit kültürlerine 0.025 mg/ml pyrimethamine verildi. Aynı kültürlere antiklastojen ajan olarak 20 uM, 40 uM ve 80 uM askorbik asit, 25 uM, 50 uM ve 100 uM folinik asit ve 40 uM askorbik asit ile 50 uM folinik asit kombine dozda verildi. Yapılan sitogenetik ve istatistiksel değerlendirmeler sonucunda pyrimethamine'nin kromozomlarda oluşturduğu aberasyonları, askorbik asit ve folinik asidin doza bağımlı olarak azalttığı belirlendi.
In this study, the effect of pyrimethamine, which is a folic acid antagonist and a antiprotozoal drug, on the formation of human chromosome aberration, anticlastogenic effect of ascorbic acid and folinic acid is researched. 18 different healthy persons, which do not use alcohol and smoking, were choosed for experimental group. 0.025 mg / ml pyrimethamine was given into the peripheric blood lymphocyte cultures which were prefere from this persons. After that 20 uM, 40 uM, 80 uM anticlastogenic agent which is ascorbic acid and 25 uM, 50 uM, 100 uM folinic acid and also 40 uM ascorbic acid and 50 uM folinic acid combine dose added into the cultures. After the cytogenetic and statistical evaluations, it is determined that, the dependend on dose of ascorbic acid and folinic acid reduced the chromosome aberration formed by pyrimethamine.
