Abstract
Bu çalışmada Bursa bölgesinde yetiştirilen Siyah Alaca ve Esmer ırkı sığırlarından toplam 136 hayvan myostatin geninin 3. ekzon bölgesinin 938. nükleotidindeki G A transisyonu ve prolaktin geninin 4. ekzonun 8398. nükleotidindeki bulunan A G transisyonu bakımından, sırasıyla BstF5I ve RsaI restriksiyon enzimleri kullanılarak, PCR-RFLP yöntemi ile incelenmiştir. Yapılan laboratuvar çalışmaları sonucunda myostatin geninin ekzon 3 bölgesinde herhangi bir G A transisyonuna rastlanılmazken, prolaktin genine ait üç genotip (AA, AG ve GG) ve iki allel (A ve G) belirlenmiştir. AG genotipinin frekansı Siyah Alaca ve Esmer sığır ırkı populasyonlarında, sırasıyla 0,2408 ve 0,4550 olarak, GG genotipinin frekansı ise Siyah Alaca sığır ırkı için 0,7396 olarak hesaplanırken, Esmer sığır ırkında 0,4225 olarak hesaplanmıştır.
In this study a total of 136 animal from Holstein Friesian cattle and Brown Swiss cattle breed rared in Bursa region were examined for exon 3 region of myostatin gene G A transisyon at 938 nucleotid position and exon 4 of Prolaktin gene A G transisyon at 8398 nucleotid position by using BstF5I and Rsal restriction enzimes, respectively with PCR-RFLP method. While the G A transisyon exon 3 region of myostatin gene was not identified, three genotypes (AA, AG and GG) and two allel (A and G) of prolactin gene were observed according to laboratory studies. Frequency of AG genotype was estimated as 0,2408 and 0,4550 for Holstein Friesian cattle and Brown Swiss cattle populations, respectively. Frequency of GG genotype was estimated as 0,7396 and 0,4225 for Holstein Friesian cattle and Brown Swiss cattle populations, respectively.
