Abstract
Bu çalışma Muğla ili Fethiye ilçesinde fasulyede hastalıklara neden olan önemli bazı virüslerin durumunu ortaya koymak amacıyla gerçekleştirilmiştir. 2012 yılı survey çalışmalarında virüs simptomu gösteren bitkilerden toplam 112 yaprak örneği alınmıştır. Hastalık etmenlerinin tanılanması serolojik yöntemler kullanılarak yapılmıştır. Örneklerin serolojik testlenmesinde Fasulye adi mozaik virüsü (Bean common mosaic virus; BCMV), Yonca mozaik virüsü (Alfalfa mosaic virus; AMV) ve Hıyar mozaik virüsü (Cucumber mosaic virus; CMV)’ne spesifik antiserumlar kullanılmıştır. DASELISA (Double Antibody Sandwich Enzyme Linked Immunosorbent Assay) testi sonucunda 112 örnekten 28’inde (%25) virüs enfeksiyonu tespit edilmiştir. Testlenen yaprak örneklerinde bu virüslerin tek başına ve karışık enfeksiyonları saptanmıştır. Enfeksiyon oranları %17.85 (20 örnek) BCMV, %3.57 (4 örnek) AMV ve %1.78 (2 örnek) CMV olarak belirlenirken karışık enfeksiyonlarda %0.89 (1 örnek) AMV+CMV ve %0.89 (1 örnek) BCMV+CMV olarak tespit edilmiştir.
This study was conducted to clarify the status of important viruses causing diseases on bean in Muğla-Fethiye subprovince. A total of 112 leaf samples were taken from plants showing disease symptoms during field surveys in 2012. Virus identification was carried out by serological methods. Specific antiserums were used for serological tests to detect Bean common mosaic virus (BCMV), Alfalfa mosaic virus (AMV) and Cucumber mosaic virus (CMV). As a result of DASELISA (Double Antibody Sandwich Enzyme Linked Immunosorbent Assay), 28 out of 112 samples (25 %) were infected with viruses. Single and mixed infections of the viruses were found according to ELISA results. Infection percentages of viruses were 17.85 % (20 samples) for BCMV, 3.57% (4 samples) for AMV and 1.78% (2 samples) for CMV. Mixed infections were about 0.89% (1 sample) for AMV+CMV and 0.89% (1 sample) for BCMV+CMV.
