Item Balık ve yetiştirme suyu kökenli Aeromonas izolatlarının antimikrobiyal duyarlılıklarının saptanması(Ankara Üniversitesi, 2017) Onuk, Ertan Emek; Yeliz, Tanrıverdi Çaycı; Çoban, Ahmet Yılmaz; Çiftçi, Alper; Balta, Fikri; Didinen, Behire Işıl; Altun, Soner; Uludağ Üniversitesi/Veteriner Fakültesi/Su Ürünleri Hastalıkları Anabilim Dalı.; 0000-0001-9096-875X; AAG-8518-2021; 56269221600Aeromonas türleri sucul ortamda yaygın olarak bulunan ve hem soğuk kanlı hayvanlarda hem de insanlar da çeşitli hastalıklara neden olabilen Gram negatif basillerdir. Bu çalışma, ülkemizde balık ve yetiştirme sularından izole edilmiş olan Aeromonas izolatlarının antimikrobiyal duyarlılık profillerinin belirlenmesi amacıyla gerçekleştirildi. Çalışmaya üç farklı coğrafik bölgeden (Karadeniz, Akdeniz, Ege) izole edilmiş olan toplam 45 Aeromonas izolatı (20 A. sobria, 10 A. hydrophila, dokuz A. salmonicida, dört A. bestiarum, iki A. veroni) dahil edildi. İzolatların 13 farklı antimikrobiyal ajana karşı antimikrobiyal aktiviteleri disk difüzyon yöntemiyle değerlendirildi. Test edilen antimikrobiyaller içerisinde gentamisin bütün izolatların duyarlı olduğu tek antimikrobiyal olarak belirlendi. Florfenikol ve siprofloksasin’in yüksek düzeyde etkin oldukları (direnç oranı %8.9), en düşük duyarlılık oranın ise amoksisilin (%17.8), ampisilin (%22.2) ve oksolonik asit’e (%35.6) karşı şekillendiği belirlendi. Conventional microbiological and API 20 E tests were used to determine phenotypic characteristics of Y. ruckeri isolates and it was determined that the Y. ruckeri showed significantly homogenous profile by microagglutination test performed by using serotype 1 and serotype 2 immunsera, and 11 of 15 isolates were serotyped as serogroup 1 while the rest 4 were serotyped as serogroup 2. In Random Amplified Polymorphic DNA (RAPD) analysis, Y. ruckeri isolates were genotyped within 2 separate clusters according to 70% similarity coefficient index and it was detected that first cluster includes 2 genotypes (YR1 and YR2) and second cluster includes 3 genotypes (YR3, YR4 and YR5). Furthermore, antibiotic resistance profiles of Y. ruckeri strains were determined and it was found that they were resistance to florfenicol, erythromycin, oxytetracycline and trimethoprim-sulphamethoxazole which have been licensed for fish health in Turkey. The aim of this study was to develop and validate the glycoprotein-based fish vaccine against Aeromonas hydrophila. For this aim, after identification and characterization of A. hydrophila isolates from fish farms, one A. hydrophila isolate was selected as vaccine strain. Antigenic glycoproteins of this vaccine strain were determined by Western blotting and glycan detection kit. The connection types of these glycoproteins were examined by glycoprotein differentiation kit. Two glycoproteins, molecular weights of 19 and 38 kDa, with SNA connection type were selected for use in vaccination trials. After their purification by SNA-specific lectin and size-exclusion chromatography, protection studies with purified proteins were performed. For challenge trials, four experimental fish groups were designated: Group I (with montanide), Group II (with montanide and ginseng), Group III [with Al (OH) 3] and Group IV [with Al(OH) 3 and ginseng]. The survival ratings of fish were determined, and protection was calculated as 21.56%, 29.41%, 69.83% and 78.88% in groups I, II, III and IV, respectively. A total of 60 bacterial isolates from water and rainbow trout samples from Turkish coastal regions (Black Sea, Aegean Sea and Mediterranean Sea) were used for 16S rDNA-Restriction Fragment Length Polymorphisms (RFLP) analysis with AluI, MboI, NarI and PstI as restriction enzymes and molecular identification was eventually completed with PCR amplification of the fstA, A. salmonicida specific gene. In addition, the presence of 6 virulence genes (aer, ser, lip, gcat, nuc and laf) in the isolates was investigated using rapid hexaplex-PCR. The frequencies of the identified Aeromonas species were 38.33% for A. sobria, 23.33% for A. hydrophila, 10.0% for A. veronii and 26.67% for A. salmonicida and only one strain was not clearly identified. The more frequent virulence genes were aer, ser, lip and gcat found in 66.66%, 61.67%, 50% and 33.33% of isolates, respectively and 14 different virulence profiles were determined. This study shows that the 16S rDNA RFLP analysis combined with fstA gene specific PCR was useful for rapid and reliable identification of Aeromonas strains and the coupled determination of the virulence genes may help to both control fish disease arising from Aeromonas spp. and alert human health risk.Item Phenotypic and molecular characterization of Yersinia ruckeri isolates from Rainbow Trout (Oncorhynchus mykiss, Walbaum, 1792) in Turkey(Schluetersche Verlagsgesellschaft, 2011) Onuk, Ertan Emek; Çiftçi, Alper; Çiftçi, Gülay; Fındık, Arzu; Balta, Fikri; Özer, Selmin; Çoban, Ahmet Yılmaz; Altun, Soner; Uludağ Üniversitesi/Veterinerlik Fakültesi/Mikrobiyoloji Anabilim Dalı.; 0000-0001-9096-875X; AAG-8518-2021; 56269221600The aim of the study was the phenotypic and molecular characterization of Yersinia (Y.) ruckeri strains, the causative agent of Enteric Red mouth Disease (ERM), by antibiotyping, random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR) and sodium dodecylsulphate-polyacrylamide gel electrophoresis (SDS-PAGE) patterns of whole cell proteins. For this aim, a total of 97 Y ruckeri isolates were analyzed. The isolates were distinguished into ten antibiotypes and six phenotypes according to their resistance properties and whole cell protein profiles, respectively. Also, a glycoprotein band of approximately 25.5 kDa was observed in all Y ruckeri strains tested. In all strains, six different RAPD types were observed. Rapid 32 STREP and conventional microbiologic tests were used for determining phenotipic features of L. garvieae strains. Altough there are differences, in Rapid 32 STREP system, between strains in terms of beta-Glucuronidase, D-ribose, sorbitol, lactose, raffinose, alanyl-phenylalanyl-proline arylamidase, pyrrolidonyl arylamidase, hippurate hydrolysis, urease tests, all strains have been confirmed as L. garvieae by API web. In RAPD PCR analysis, in which ERIC 2 primer is used, L. garvieae isolates were genotyped within 3 separate clusters according to similarity coefficient index of 70%, and it was detected that a vast majority of isolates with Turkey-origin (8 isolates) belongs to predominant type LG1 genotype. Tıp ve veterinerlik alanında yaygın olarak kullanılan kinolonların çevresel ortamlarda değişmeden kalabilme özelliği, dirençli mutantların seçiminde önemli rol oynamaktadır. Plazmid aracılı direnç, kinolonlara karşı direncin gelişiminde etkin olan mekanizmalardan biri olup, qnr, qepA, aac(6’)-Ib-cr ve oqxAB genleri direnç determinantları olarak tanımlanmıştır. Birçok farklı qnr determinantının, başta Enterobacteriaceae ailesi olmak üzere çeşitli bakterilerde saptanması, bu yapıların doğada bulunabileceğini vurgulamaktadır. Yakın zamanda sudan izole edilmiş Aeromonas spp. izolatlarında plazmid aracılı kinolon direnci tespit edilmiştir. Bu çalışmanın amacı, ülkemizde sukaynaklı Aeromonas suşlarında qnr gen varlığının araştırılmasıdır. Çalışmaya, Türkiye’nin üç farklı coğrafi bölgesindeki (Ege, Akdeniz ve Karadeniz) balık ve sulardan izole edilmiş toplam 45 Aeromonas spp. izolatı dahil edilmiştir. İzolatların tür düzeyinde tanımlanması 16S rDNA-RFLP (Restriction fragment length polymorphism) ve multipleks polimeraz zincir reaksiyonu (M-PCR) ile yapılmış ve 20 suş A.sobria, 10 suş A.hydrophilia, 9 suş A.salmonicida, dört suş A.bestiarum ve iki suş A.veronii olarak tanımlanmıştır. Plazmid aracılı kinolon direnç genlerinden qnrA, qnrB, qnrC ve qnrS genlerinin varlığı M-PCR ile araştırılmış; qnr pozitif olarak saptanan dokuz izolata dizi analizi uygulanmıştır. Dizi analizi sonucuna göre, altı A.sobria, iki A.bestiarum ve bir A.hydrophila izolatında (9/45; %20) qnr varlığı tespit edilmiş; bu gen GenBank veri tabanı ile karşılaştırıldığında qnrS2 olarak tanımlanmıştır. Tüm qnrS2 pozitif Aeromonas izolatları siprofloksasine duyarlı olarak saptanırken, beş izolat nalidik aside dirençli bulunmuştur. Bu çalışma, ülkemizde su kaynaklı Aeromonas türlerinde plazmid aracılı kinolon direncinin araştırıldığı ve qnrS2 varlığının tespit edildiği ilk çalışma olma özelliğini taşımaktadır. Sonuç olarak, su kaynaklı mikroorganizmalardaki farklı direnç genlerinin diğer bakterilere ve klinik izolatlara aktarılmasının, enfeksiyonların tedavisinde potansiyel bir risk oluşturacağı düşünülmüştür.
