In the study, a total of 139 healthy cows with the serum progesterone > 8 ng/ml were used as animal material. Estrus synchronization protocol was applied and cows were divided into three different groups as conventional semen group (G1, n=46), sexed semen group (G2, n=47), and embryo transfer group (G3, n=46) considering age, body condition score, lactation number, and body weight. Cows in G1 and G2 were inseminated with conventional semen and sexed-semen, respectively. Embryo transfer was performed to cows in Group 3 (n=46) 7th day after estrus. The embryos were transferred to recipients as freshly. Pregnancy examinations for the cows were conducted on the 30th and 60th days of gestation. The statistical analysis of the obtained results was performed. Pregnancy rates were detected as %50, %46,8 and %69,56 in G1, G2, and G3 respectively on 30th day. On 60th day, however, pregnancy rates were %45.60, %42.55 and %67.39 in G1, G2, and G3 respectively due to embryonic losses. The pregnancy rate was significantly higher in the embryo transfer group (G3) compared to the conventional semen (G1) and sexed-semen (G2) group (p<0.05) on both 30th and 60th days. There was no significant difference between G1 and G2 (p>0.05). Findings led to the conclusion that higher serum progesterone level may increase pregnancy rate in cattle. A total of 16 dogs with locally advanced mammary tumors aged 8-14 years was used in this study. Samples were taken from all the dogs via Tru-Cut biopsy and fine needle aspiration biopsy of the lymph nodes prior to neoadjuvant chemotherapy. The samples were sent to the pathology laboratory for histopathological examination. Based on the histopathology results, mammary tumors were diagnosed as adenocarcinoma (n=13), tubulopapillary carcinoma (n=2) and malignant mixed tumors (n=1). For the neoadjuvant chemotherapy, doxorubicin (25-30 mg/m(2)) and cyclophosphamide (100 mg/m(2)) were administered via slow IV injection 4 times at 3-week intervals. Clinical findings such as fever, vomiting, anorexia, lethargy, weight loss, alopecia, enteritis, hematuria, and skin ulceration and laboratory findings such as a decrease in neutropenia and hematocrit values were observed. Based on the longest diameter of the tumors before and after four weeks of chemotherapy, a complete response was detected in 2 dogs (12.5%), a partial response was observed in 9 dogs (56.25%) and the disease was stable in 5 dogs (31.25%). Semen samples were collected from eight Saanen buck, and each semen sample was split into four equal aliquots and diluted with different concentrations of PRP supplemented extenders [no PRP (control), 0.5 x 107/ml PRP, 1 x 107/ml PRP, or 2 x 107/ml PRP]. Motility, plasma membrane functional integrity, acrosome integrity, mitochondrial membrane potential, DNA integrity and malondialdehyde concentrations (MDA) were measured and analyzed at post-thaw. The results showed that 2 x 107/ml PRP group had a positive effect on motility (62.41 +/- 4.24), membrane functional integrity (71.11 +/- 2.90), mitochondrial membrane potential (69.70 +/- 1.99), DNA integrity (7.22 +/- 0.93) and MDA levels (2.56 +/- 0.73) at post-thaw (P < 0.05). The results of the study demonstrated that autologous PRP has a protective effect on cryopreservation of buck spermatozoa and the fertility effects are worthy of further study.Item Siyah alaca sütçü düvelerde vitamin D3 uygulamalarının kan serumu anti-müllerin hormon seviyesi üzerine etkilerinin araştırılması(Bursa Uludağ Üniversitesi, 2022-05-11) Koca, Davut; Nak, Yavuz; Bursa Uludağ Üniversitesi/Sağlık Bilimleri Enstitüsü/Veteriner Fakültesi/Doğum ve Jinekoloji Anabilim Dalı.; 0000-0002-7962-6959AMH düzeyini etkileyen çeşitli faktörler bulunmaktadır. Vitamin D, AMH düzeyini etkileyen faktörlerden sadece biridir. Başta insanlar olmak üzere yapılan çok sayıda in vivo ve in vitro çalışmada, Vitamin D’nin AMH düzeyini doğrudan veya dolaylı olarak etkilediği bildirilmektedir. Ancak düvelerde Vitamin D’nin serum AMH düzeyleri üzerine etkilerinin incelendiği bir araştırma bulunmamaktadır. Bu çalışmada tohumlanmamış 11-13 aylık sütçü düvelerde oral ve kas içi Vitamin D3 uygulamalarının AMH ve 25(OH)D düzeyleri üzerine etkileri araştırılmıştır. Çalışmada kullanılan hayvan materyali, oral (Grup1, n=20), kas içi (Grup2, n=20) ve kontrol grubu (Grup3, n=20) olmak üzere 3 gruba ayrılmıştır. Oral gruba ve kas içi gruba tek seferde 5 milyon IU Vitamin D3 uygulaması yapılmış, kontrol grubuna ise Vitamin D3 uygulaması yapılmamıştır. Kan örnekleri uygulama öncesi (0. gün) ve uygulamayı izleyen 7, 14 ve 28. günlerde toplanmıştır. AMH konsantrasyonları kan serumunda elektrokemilüminesans yöntemiyle, 25(OH)D konsantrasyonları ise kemilüminesan immunolojik test yöntemiyle belirlenmiştir. Çalışma sonucunda oral ve kas içi vitamin D3 uygulamasının 25(OH)D konsantrasyonunu 7, 14 ve 28. günlerde uygulama öncesine göre önemli düzeyde artırdığı belirlenmiştir (p<0.05). Oral Vitamin D3 uygulaması sonrası 25(OH)D düzeyinin 7. günde pik seviyeye ulaştığı, 14 ve 28. günlerde ise azaldığı tespit edilmiştir (p<0.05). Kas içi uygulamada ise 25(OH)D düzeyinin, uygulama öncesine göre sürekli artış gösterdiği ve 28. günde pik düzeye ulaştığı belirlenmiştir (p<0.05). Oral ve kontrol grubunda uygulama öncesine göre 7, 14 ve 28. günlerdeki AMH düzeyi değişimlerinin istatistiki açıdan önemsiz olduğu tespit edilmiştir (p>0.05). Kas içi grupta ise AMH düzeyinin 7. ve 28. günlerde uygulama öncesine göre istatistiki açıdan daha yüksek olduğu belirlenmiştir (p<0.05). Kontrol grubunda 0. güne kıyasla AMH düzeylerinde azalma gözlemlenmiştir. Elde edilen bulgulara dayanarak, Vitamin D3’ün veriliş yolunun 25(OH)D düzeyini etkilediği, oral grupta 7. günde pik düzeye ulaşırken, kas içi grupta sürekli bir artış sağladığı belirlenmiştir. Vitamin D3 uygulanan gruplarda AMH konsantrasyonunda artışlar gözlenmiş ve kontrol grubunda gözlenen AMH konsantrasyonlardaki dalgalanmalardan farklı olarak düzenli bir seyir izlediği tespit edilmiştir.
