Antioxidant activity and total phenolic contents were evaluated in acetone, ethanol, and methanol extracts of these three species. Antioxidant activity was measured by ABTS [2,2'-azino-bis(3-ethylbenzothiazoline- 6-sulphonic acid)] method and total phenolic contents were measured by Folin-Ciocalteu method. High-performance liquid chromatography (HPLC) was used for the determination of lichen acids. It can be concluded from stictic and usnic acids contents that the order of solvent efficiency is acetone > ethanol > methanol and acetone > methanol > ethanol, respectively. Broth microdilution method was performed to determine the minimum inhibitory concentration (MIC) of the methanol extracts of three Usnea species. The MIC values of all the extracts ranged from 64 mu g/mL to 512 mu g/mL for all the bacterial strains that were tested in this study, and all the Fluoro quinolone-resistant Escherichia coli isolates (except for E101) were sensitive to the methanol extracts of the three Usnea species. This paper is the first study to determine the stictic acid content in U. intermedia and U. filipendula. The mercury was converted to metallic vapour in a reaction cell by tin chloride solution, swept by nitrogen into a gold-plated graphite tube and subsequently atomized. The influence of four experimental factors on the sensitivity of mercury determination was examined using two level factorial designs. It was found that the solution volume is the most significant parameter.Item Bazı enzimleri kullanarak biyodizel üretimi ve biyodizel özelliklerinin analitik metotlarla araştırılması(Uludağ Üniversitesi, 2008) Yücel, Yasin; Demir, Cevdet; Uludağ Üniversitesi/Fen Bilimleri Enstitüsü/Kimya Anabilim Dalı.Tükenen petrol kaynakları ve buna bağlı enerji krizleri nedeniyle son yıllarda alternatif enerji kaynakları önemli araştırma alanı haline gelmiştir. Biyodizel, yenilenebilen, sera gazlarını azaltıcı düşük emisyona sahip, biyolojik olarak parçalanabilen ve zehirli olmayan yakıt olması nedeniyle petrol esaslı yakıtlara alternatif bir yakıttır. Biyodizel trigliseridlerin metanol ile transesterleşmesiyle elde edilen yağ asidi metil esterleri olarak tanımlanabilir. Son zamanlarda yağ asidi metil esterlerinin saf olarak elde edilmesi, gliserolün yan ürün olarak kolaylıkla ayrılması gibi nedenlerle biyodizel yakıt üretiminde lipaz kullanılarak enzimatik transesterleşme metodu çok daha cazip hale gelmiştir. Fakat yüksek enzim maliyetleri endüstriyel uygulamalar için dezavantajdır. Üretim maliyetinin düşürülmesi için enzim uygun bir taşıyıcı üzerine tutturulabilmeli ve tekrar kullanılabilmelidir.Bu çalışmada, Candida antarctica (Novozym 435), Thermomyces lanuginosus (Lipozyme TL-IM) ve Rhizomucor miehei (Lipozyme RM-IM) immobilize lipazlarının metanol beraberinde kanola yağını transesterleşme reaksiyonu araştırıldı. Reaksiyon süresi, sıcaklık, enzim miktarı ve metanol-yağ molar oranı gibi kanola yağını transesterleşme reaksiyonu ile metil esterlerine dönüştürmede etkin olan parametreler merkezi kompozit dizayn ve yüzey analizleri kullanılarak incelendi.Transesterleşme reaksiyonunda optimum koşullar, yağ ağırlığının (4%-21%)' i oranında enzim, (1:1,6-1:4,1) yağ-alkol molar oranı, (34ºC-55ºC) reaksiyon sıcaklığı ve (4-6) saat reaksiyon süresi olarak belirlendi. En yüksek biyodizel dönüşümü (%97,2) enzim karışımı kullanılarak elde edildi.Enzimlerin immobilizasyonunda kullanılmak üzere mikro gözenekli polimer matriks stiren divinil benzen kopolimerinden sentezlendi. Lipozyme TL-100L ve Novozyme 388 lipazları celite 545, silikajel ve stiren divinil benzen kopolimerleri üzerine absorpsiyon ve kovalent bağlama metotları kullanılarak immobilize edildi. Enzimlerin immobilizasyon verimliliği ve aktiviteleri belirlendi. Destek maddeleri ve immobilize edilmiş türevleri taramalı elektron mikroskobu ve fourier transform infrared spektrometresi (FTIR) ile incelendi.Biyodizel üretimi kanola yağı ve metanol karışımının bir peristaltik pompa yardımıyla mikro gözenekli polimer kolon içinden 24 saat sürede devridaimi ile yapıldı. Gaz kromatografi analiz sonuçlarına göre en yüksek metil ester dönüşümü %97 olarak hesaplandı. Tekrar kullanım açısından mikro gözenekli polimer üzerine tutturulan lipaz 10 kez kullanılmasına rağmen önemli bir aktivite kaybı gözlenmedi.Item Bitkisel atık yağların karakterizasyonu ve biyodizel üretiminde değerlendirilmesi(Uludağ Üniversitesi, 2010-01-11) Aybastıer, Önder; Demir, Cevdet; Uludağ Üniversitesi/Fen Bilimleri Enstitüsü/Kimya Anabilim Dalı.Thermomyces lanuginosus lipaz enzimi mikroporoz stiren-divinilbenzen- poliglutaraldehit (STR-DVB-PGA) polimerine kovalent olarak immobilize edilmiştir. Merkezi kompozit dizayn ile immobilizasyon üzerine enzim konsantrasyonunun (% 4-16 v/v), tampon pH'nın (6,0-8,0), tampon konsantrasyonunun (20-100 mM), immobilizasyon süresinin (8-40 saat) etkisi incelenmiş ve etkili faktörler optimize edilmiştir. Enzim konsantrasyonu, pH ve tampon konsantrasyonunun spesifik aktivite üzerinde etkili olan önemli faktörler olduğunu bulunmuştur. Optimum koşullardaki tahmini spesifik aktivite 8.78 ?mol pNP/mg enzim.dk olarak hesaplanmış, deneysel spesifik aktivite ise 8,41 ?mol pNP/mg enzim.dk olarak belirlenmiştir. Metanol ve t-bütonolün immobilize lipazın spresifik aktivitesi üzerine etkisi incelenmiştir. İmmobilize lipazın aktivitesi t-bütanolde hemen hemen değişmezken, metanolde büyük oranda azalmaktadır. Çalışmanın ikinci aşamasında çapraz bağlayıcı olarak glutaraldehit kullanılarak kitosan küreler hazırlanmıştır. Candida antarctica A lipaz enzimi kitosan küreler üzerine kovalent olarak immobilize edilmiştir. Bu küreler üzerine yapılan immobilizasyonda en yüksek spesifik aktivite, % 12 (v/v) enzim konsantrasyonunda ve 24 saat immobilizasyon süresinde elde edilmiştir. Optimum koşullarda enzim immobilize edilen STR-DVB-PGA polimeri ve kitosan küreleri kullanılarak ön işlem uygulanmamış atık kanola yağından biyodizel üretilmiştir. Her iki destek maddesi kullanılarak üretilen biyodizel verimleri sırası ile % 85,95 ve % 59,32 dir. STR-DVB-PGA destek maddesi ile daha yüksek oranda biyodizel elde edildiği görülmüştür.Item Bursa'da yetiştirilen bazı ahududu (Rubus idaeus L.) ve böğürtlen (Rubus fruticosus L.) çeşitlerinin fenolik bileşiklerinin sıvı kromatografisi kütle spektrometresi (LC-MS) ile incelenmesi ve antioksidan aktivite tayinleri(Uludağ Üniversitesi, 2009-07-21) Sarıburun, Esra; Demir, Cevdet; Uludağ Üniversitesi/Fen Bilimleri Enstitüsü/Kimya Anabilim Dalı.Bursa da yetiştirilen beş ahududu (Aksu kırmızısı, Rubin, Newburgh, Hollanda boduru, Heritage) ve dört böğürtlen (Bursa 1, Bursa 2, Chester, Jumbo) çeşidinin fenolik madde içerikleri taze, ilk dondurulmuş ve altı ay süreyle -220C de muhafaza edildikten sonra LC-MS/MS ile tayin edilmiştir. Ellagik asit ahududu ve böğürtlen türlerinin temel fenolik maddesi olduğu tesbit edilmiştir. Ellagik asit (1350,36-727,90 mg/kg taze meyve), feruik asit (820,78-338,27 mg/kg taze meyve), kafeik asit (754,85-202,78 mg/kg taze meyve), para-kumarik asit (361,68-142,63 mg/kg taze meyve), para-hidroksibenzoik asit (534,20-233,29 mg/kg taze meyve) and kuersetin (46,97-27,31 mg/kg taze meyve) ahududu çeşitlerinin, ellagik asit (1828,07-1555,13 mg/kg taze meyve), ferulik asit (757,69-413,82 mg/kg taze meyve), kafeik asit (736,85-337,89 mg/kg taze meyve), para-kumarik asit (877,45-287,15 mg/kg taze meyve) ve kuersetin (74,69-56,78 mg/kg taze meyve) böğürtlen çeşitlerinin su ekstraklarında tayin edilmiştir. Ahududu çeşitlerinin fenolik bileşik içerikleri farklılıkları böğürtlen çeşitlerinden daha fazladır. Altı ay süreyle -220C de muhaza edilen ahududu ve böğürtlen çeşitlerinin fenolik bileşik içeriklerinin başlangıç miktarlarına göre ortalama iki kat azaldığı belirlenmiştir.İkinci olarak ahududu ve böğürtlen türlerinin toplam fenol, toplam flavonoid, toplam antosiyanin, askorbik asit miktarları ile ABTS, DPPH ve CUPRAC metotları ile antioksidan aktivite değerleri UV-VIS spektrofotometresi ile tayin edilmiştir. Ahududu ve böğürtlen çeşitleri ABTS, DPPH, CUPRAC, toplam fenol ve toplam flavonoid içerikleri metanol ekstraklarında, toplam antosiyanin ve askorbik asit içerikleri ise su ekstraklarında yüksek değerlere sahiptir. Ahududu ve böğürtlen çeşitlerinin antioksidan aktivitelerinin çeşitlerin fenolik içerikleriyle direk olarak ilişkili olduğu bulunmuştur. Antioksidan aktivite ile toplam fenol, toplam flavonoid ya da askorbik asit arasında doğrusal ilişki olduğu gözlenmiştir. Elde edilen sonuçlar ahududu ve böğürtlen türlerinin zengin antioksidan kaynağı olduğunu göstermektedir.Item Ceviz türleri (pikan ve Türk cevizi) yağlarının oksidasyon özelliklerinin araştırılması, yağ asitleri ve tokoferol tayinleri(Uludağ Üniversitesi, 1990-09) Demir, Cevdet; Çetin, Mehmet; Uludağ Üniversitesi/Fen Bilimleri Enstitüsü/Kimya Anabilim Dalı.Bu çalışmada, pikan ve Türk cevizi meyvelerinden pres ve çözücü ekstraksiyonu (izopropanol ve hekzan) yöntemiyle elde edildi. Bu yağların yağ asitleri ve tokoferol analizleri yapıldı, yağların oksidasyon özellikleri araştırıldı. Bulunan sonuçlar ayçiçek yağı ile kıyaslandı. Yağ asitleri analizi sonunda Türk cevizi yağında doymamış yağ asitleri oranı yüksek, pikan ve ayçiçek yağında daha düşük olduğu bulundu. HPLC yöntemiyle yapılan tokoferol tayinleri sonunda pikan cevizi yağın da sadece y- tokoferol, Türk cevizi yağında < -, T - ve 6- tokoferol, ayçiçek yağında ise ve -t tokferol bulunmuştur. Çözücü ekstraksiyonu ile elde edilen yağlarda tokoferol miktarının daha düşük olduğu görülmüştür. Bu yağların indüksiyon periyotları bulunarak oksidasyona karşı kararlılıkları araştırıldı. A fuzzy rule-building expert system (FuRES), support vector machine classification tree (SVMTreeG), and super partial least-square discriminant analysis (sPLS-DA) were used to develop classification models. Normalization, retention time alignment, square root transform, and dissimilarity kernel were evaluated as data preprocessing methods. The bootstrapped Latin partition was used with 100 bootstraps and 4 partitions. Classification rates of FuRES and SVMTreeG with a square root transform were 97.6 +/- 0.4% and 97.6 +/- 0.4% for classifying the type of honey, respectively. The measures of precision are 95% confidence intervals. Determination of heavy metals such as Cu, Pb, Ag, Ni, Mn, Cd, Zn, Fe, Co and Mg in original marbles have been carried out by atomic absorption spectrophotometer, and Na and K by atomic emission spectrophotometer. Information about geographical origin is often required of marble samples. In order to obtain such information, principal component analysis, correlation analysis and cluster analysis techniques can be used as valid tools for processing series of data from chemical analysis. The techniques were applied to the raw and standardised data, showing different clusters related to the origin of marble samples. Some predictions about the origin of samples are made and showing the potential of chemometric techniques and their possible contribution to the characterisation of marbles used in historical buildings.Item Cold vapour generation and on-line trapping of cadmium species on quartz surface prior to detection by atomic absorption spectrometry(Royal Soc Chemistry, 2005) Korkmaz, Deniz; Aydın, Fırat; Ataman, O. The cold vapour generation technique was used for the generation of Cd species. The trapping medium was formed by external heating of the inlet arm of a quartz T-tube. The generated analyte species were trapped on a quartz surface heated to the collection temperature, 350 degreesC, and the collected species were revolatilized when the trap was heated further to revolatilization temperature, 1000 degreesC, and hydrogen gas was introduced in the trapping medium. Two-level fractional factorial design and central composite design were used to optimize generation conditions in the flow injection mode. The results of the fractional factorial design demonstrated that the factors and their interactions were statistically significant. Three factors, length of reaction coil, carrier HCl concentration and NaBH4 concentration, were considered to be the most significant parameters in the optimization and their optimum values were found to be 30 cm, 0.3 M and 3% (m/v), respectively. Sea-water (BCR), tomato leaves (NIST 1573a) and oyster tissue ( NIST 1566b) standard reference materials were analyzed to assess the accuracy of the proposed method. For a collection period of 3.0 min, i.e., 6.0 ml sample volume, the 3sigma limit of detection was 1.8 pg ml(-1); the enhancement factor for LOD was found to be 90 as compared with FI-HGAAS. The dyes assessed represent low (CI Disperse Blue 56), medium (CI Disperse Blue 60) and high (CI Disperse Blue 79) energy level dyes. UV-visible spectroscopy, HPLC-DAD and COD analysis were performed to follow the removal of color from effluent. The results indicated that color removal rates up to 100% and COD removal rates up to 68% could be achieved via ozonation. HPLC analysis and UV-vis analysis were performed for all of the dyes, and a degradation mechanism was proposed for CI Disperse Blue 79.Item Deri boyaması üzerine etki eden analitik parametrelerin optimizasyonu(Uludağ Üniversitesi, 2004-03-26) Yıldırım, Nurhayat; Demir, Cevdet; Uludağ Üniversitesi/Fen Bilimleri Enstitüsü/Kimya Ana Bilim Dalı.Deri endüstrisinde boyama, tabaklama ve bitirme işlemleri arasmda yer alan bir geçiş prosesidir. Deri boyacılığı; derinin sabit bir kimyasal yapıya sahip olmaması ve etkili birçok parametrenin bulunması sebebiyle, tekstil boyacılığına nazaran oldukça karmaşıktır. Boyarmaddenin üç boyutlu kollagen lifleri arasına bağlanması diğer materyallerden farklı oltıp, birbirini takip eden birçok olayın bir sonueudur. Tabaklama sürecinde uygun boyama metodu seçilirken, boyama şartlarının optimize edilmesi, boyamanın homojen yapılması, partiler arasında farklılık bulunmaması dikkate alınır ve sonuçta rasyonel ve ekonomik bir çalışma mümkün olur. Çalışmamızda; deri endüstrisinde boyamaya etki eden analitik parametrelexin optimizasyon Çalışmaları yapılmıştır. Bu amaçla seçilen boyalarla (CI. Aftiâ Yellow 194, CX Acid Red 357, CI. Acid Blue 193, CX Acid Brown 354, CX Acid Black 210) boyama çalışmaları yapıldı ve her bir boya için, boyama ortamına ilişkin secileri altı parametre incelendi Bu parametreler; banyo (boyana ortamındaki su) miktarı, sıcaklık, süre, pH, formik asit miktarı, yardımcı madde miktarıdır. Seçilen faktörlerin boyamaya olan etkileri kemomeîrik yöntemlerle araştırıldı. Bu amaçla her bir boyarmadde için Plackett - Burman Deneysel Dizayn yöntemi ile en etkili parametrelerin süre ve yardımcı madde miktarı olduğu tespit edildi Bunların optimum koşuHannın belirlenmesi amacryla Doeblert Deneysel Dizaynı uygulandı ve her bir boyarmadde için optimum koşullar belirlendi Yapılan boyama işlemleri sonucunda, boyama ortamında bağlanmadan kalan boyarmadde miktarımn tespiti için ekstraksiyoB çalışmaları yapıldı ve UV- Vis spektroskopiyle absorbans ölçümleri yapıldı. High-performance liquid chromatography and spectrophotometric approaches were used to determine the antioxidant capacities of fruit juices. The importance of calibration design was investigated by calculating the prediction and validation errors. The influences of using independent validation sets were emphasized. Calibration design is shown to have major effect on principal component regression and partial least squares errors. The models developed on the basis of the mean-centered data were able to predict the total antioxidant activity with a precision comparable to that of the reference [2,2-azino-di-(3-ethylbenzothialozine-sulfonic acid)] method. The partial least squares model seems preferable because of its predictive and describing abilities and good interpretability of the contribution of compounds to the antioxidant capacity. (Prunella vulgaris L., Prunella laciniata L., Prunella grand:flora L. and Prunella orientalis Bornm.) belong to the family of Lamiaceae and representing popular Western and Chinese herbal medicine were examined for the content of phenolic compounds. Phenolic acids (rosmarinic acid, caffeic acid, ferulic acid, chlorogenic acid, protocatechuic acid), flavonoids (rutin, quercetin) in different quantitative proportions depending on extracts were determined by the rapid, selective and accurate method combining solvent/acid hydrolysis extraction and high performance liquid chromatographydiode array detection (HPLC-DAD). Water, methanol, butanol, acetonitrile, ethyl acetate, hexane and their acidic solutions were used to examine the efficiency of different solvent systems for the extraction of phenolic compounds. The amounts of phenolic compounds in honeys were determined by high performance liquid chromatography-diode array detection. The antioxidant capacities were determined by ABTS and CHROMAC methods. The total phenolic content of honeys were determined by spectrophotometric method using the Folin-Ciocalteu reagent. Caffeic, protocatechuic, and p-hydroxybenzoic acids are the major phenolic compounds with the contents of 44.52, 17.48, and 21.50 mg/kg, respectively in chestnut honeys. Chestnut honeys exhibited the higher antioxidant and better antimicrobial activities, and than the floral honeys. Floral honeys contain similar amounts of propolis flavonoids such as pinocembrin, chrysin, and galangin. The results show that these flavonoids can be used as chemical markers in honey samples. Practical applicationsHoney is an important natural product that contains major compounds including glucose and fructose and minor components such as amino acids, organic acids, enzymes, vitamins, proteins, phytochemical substances mainly flavonoids and other phenolic compounds. Plants are important sources of natural compounds that contain polyphenolic derivatives such as flavonoids and phenolic acids. These bioactive compounds can be transferred from plants to honey. Polyphenolic compounds were recognized as the major components responsible for health-promoting properties of honey. This article evaluates the antimicrobial and antioxidant activities and phenolic compounds of chestnut and floral honeys. Fatty acids and tocopherols of oils were determined and oxidation stability of oils was investigated. The results were compared with sunflower oil. Determination of fatty acids was performed by gas chromatography (GG). It was observed that composition of unsaturated fatty acids is high in walnut oil. Tocopherols were determined in oils by high performance liquid chromatography HPLC. Only gamma-tocopherol was found in pecan oil, alpha- gamma- and delta-tocopherols in walnut oil, and alpha- and gamma-tocopherols in sunflower oil. It was observed that more amounts of tocopherols are obtained by pressing. Oxidative stability of oils was followed at 100 degrees C by determining the peroxide value. Central composite design (CCD) was employed to optimize the conditions for the maximum HRP activity and to understand the significance and interaction of the factors affecting the activity of immobilized HRP. The results indicated that enzyme concentration and immobilization time were significant factors for the immobilization of HRP. The optimum conditions were determined as enzyme concentration 0.25 mg mL(-1), pH 8.0 and immobilization time 20h. The recovered activity was obtained as 81% after immobilization under optimal conditions. Total phenol content was determined in four Prunella L. species (Prunella vulgaris L., Prunella laciniata (L.) L., Prunella orientalis Bornm., Prunella grandiflora L.) extracted using methanol, water and methanol/water (4 : 1, v/v). The enzymatic method is based on the spectrophometric measurement of the final quinone-imine colored product, absorbing at 510 nm, by HRP oxidation in presence of hydrogen peroxide. The results were compared with those obtained by applying the Folin method. The highest total phenol content was obtained with the methanol/water (4 : 1, v/v) extract of Prunella vulgaris L. by immobilized HRP method (63.75 mg gallic acid equivalent (GAE) per g dried plant). The method is based on the spectrophotometric measurement of the final quinone-imine colored product, absorbing at 510 nm, by horseradish peroxidase oxidation in presence of hydrogen peroxide. The effects of solvents were investigated for the extraction of phenolic compounds in Prunella L. plants. HRP was covalently attached on the STY-DVB-PGA with 90% efficiency at optimum immobilization conditions. The total phenol contents ranged from 4 to 152 mg GAE per g of dried sample for pure solvents and ranged from 19 to 763 mg GAE per g of dried sample for acidic extracts using the immobilized enzymatic method. STY-DVB-PGA beads are a good potential support for the immobilization of HRP enzyme and can be used for determination of the total phenolic content of plants. In the analysis of four Prunella L. species, high correlations were obtained between enzymatic and Folin methods, supporting the validity of the enzymatic method proposed. The enzymatic (free/immobilized) methods are more specific and rapid than the Folin method. The HRP was immobilized on the styrene-divinylbenzene-polygluteraldehyde (STY-DVB-PGA) beads. Total phenol content determined by immobilized and free enzymatic methods was compared with those obtained by applying the Folin-Ciocalteu method. The total phenol content by immobilized HRP method ranged from 254 to 973 mg gallic acid equivalent (GAE) per 100 g fw as well as 105 to 435 mg ellagic acid equivalent (EAE) per 100 g fw in raspberry and blackberry cultivars. 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical values were correlated with total phenol content in raspberry using immobilized enzymatic method as GAE (R-2 = 0.7450, P = 0.05) and EAE (R-2 = 0.7449, P = 0.05). All antioxidant activity values were highly correlated with total phenol content in blackberry using free and immobilized enzymatic methods as GAE (R-2 = 0.9777 and 0.9223, P = 0.05 respectively) and EAE (R-2 = 0.9979 and 0.9223, P = 0.05 respectively). A significant additivity was observed using individual phenolic compounds by immobilized enzymatic method as 95% (GAE) and 90% (EAE). The assay is based on the spectrophotometric measurement of colored a chelate complex of Cr(III) and diphenylcarbazone formed by the reaction of Cr(VI) and 1,5-diphenylcarbazid in acidic medium. Phenolic compounds react with excessive amounts of Cr(VI) at low pH values, causing reduction of Cr(VI) to Cr(III) and conversion of phenols to oxidized products. The assay comprises of the antioxidant with a chromium(VI) solution, a 1,5-diphenylcarbazid in acidic medium and subsequent measurement of the developed absorbance at 540 nm after 50 min. The color development is stable for phenolic compounds in plant and fruit. The selectivity of the assay for phenolic compounds was improved by adjusting pH to 2.8 and reduction potential between 0.2 and 0.9 V. The developed assay was successfully applied to the measurement of antioxidant capacity of three plants and one fruit (Prunus divaricata Ledeb.subsp. divaricata) samples and comparable results were obtained by ABTS and CUPRAC assays.Item DNA'nın oksidatif hasarı üzerine fenolik maddelerin etkisi(Uludağ Üniversitesi, 2016-04-25) Aybastıer, Önder; Demir, Cevdet; Uludağ Üniversitesi/Fen Bilimleri Enstitüsü/Kimya Anabilim Dalı.DNA'nın oksidatif baz hasarı üzerine karvakrol, timol, timokinon ve kekik bitkisinden elde edilen yağ altı suyu, uçucu yağı ve ekstraktların etkisi incelenmiştir. Öncelikle çalışmada kullanılan kekik türü olan Origanum vulgare L.'nin antioksidan özellikleri belirlenmiştir. İncelenen kekik türünün antioksidan kapasitesinin yüksek ve içerdiği temel antioksidan maddelerin karvakrol ve timol olduğu tespit edilmiştir. Sulu çözelti ortamında karvakrol, timol ve timokinon moleküllerinin Fenton reaksiyonu ile oluşturulan hidroksil radikalini süpürme etkisi incelenmiştir. Hidroksil radikalini (•OH) en hızlı timokinonun süpürdüğü belirlenmiştir. DNA oksidatif baz hasar ürünlerinin GC-MS/MS ile analizi için metot geliştirme çalışmaları yapılmıştır. Analiz öncesi türevlendirme koşulları merkezi kompozit dizayn ile optimize edilmiştir. GC-MS/MS çalışma koşulları da optimize edilerek ilk kez 18 farklı DNA oksidatif baz hasar ürünü eş zamanlı ve çok düşük konsantrasyonlarda analiz edilebilmiştir. Buzağı timüs DNA'sı antioksidan içeren ve içermeyen ortamlarda Fenton reaksiyonuna maruz bırakılıp, oluşan DNA oksidatif baz hasar ürünleri tayin edilmiştir. Antioksidan varlığında, oluşan DNA oksidatif baz hasar ürünü miktarının azaldığı görülmüştür. Aynı çalışma malign (A549) ve sağlıklı (BEAS-2B) akciğer hücreleri üzerinde de denenmiştir. Hücre kültürü ortamında çalışılan antioksidanlardan ve H2O2 eklendikten 48 saat sonra hücreler alınıp, DNA izolasyonu yapılmıştır. Elde edilen DNA'daki oksidatif baz hasar ürünleri analiz edilmiştir. Hücre kültürü ortamında da kullanılan antioksidanların DNA'yı hidroksil radikaline (•OH) karşı koruduğu belirlenmiştir. Tez kapsamında kullanılan antioksidanlardan timokinon DNA'yı oksidasyona karşı korumada daha etkili olduğu bulunmuştur.
