Glikojen tesbitinde fiksatiflerin etkisinin histolojik olarak incelenmesi

Abstract

Bu çalışmada karaciğer glikojen tesbitinde değişik fiksatiflerin etkileri, fiksasyon süresi ve ortam şartları (sıcaklık farkı, hava ve su) içinde incelenip, birbirleriyle kıyaslanması ve en iyi yöntemin saptanması amaçlandı. Karaciğerden alınan örneklerde deney sonucunda; 24 ve 48 saat oda sıcaklığında (20°C) Orth solüsyonunda veya 24 saat 44°C'de Orth solüsyonu ve tuzlu-formalinde veya 4 ve 8 gün oda sıcaklığında Orth solüsyonu ve tuzlu-formalinde fikse edilen parçalarda glikojenin en iyi şekilde korunduğu gösterildi.

In this study, the effects of various fixatives on the fixation of liver glycogen in different environmental conditions and of different fixation time was investigated and the results were compared in order to determine the best method. Experimental results showed that the glycogen was stored successfully in Orth's fluid at room temperature (20° C) for 24-48 hours or in Orth's fluid and formal-saline at 44°C for 24 hours or in Orth's fluid and formal-saline for 4-8 days in room temperature.
