Semen samples were collected from sexually mature drones and pooled. Then pooled semen was diluted with different concentrations of RTSP (2.5%/5% / 10%) and without RTSP (control) supplemented extenders. Motility, plasma membrane functionality, acrosome integrity, and mitochondrial function were negatively affected by the cryopreservation process but DNA integrity was not affected. Membrane lipid peroxidation status was also analyzed using the malondialdehyde (MDA) concentration at frozen-thawed. RTSP10 groups had a positive effect on sperm motility (64.00 +/- 3.38%), plasma membrane functionality (72.07 +/- 2.12%), acrosome integrity (86.20 +/- 2.11%), and mitochondrial function (69.33 +/- 2.94%) compared to the control group (P< 0.05). The study shows that RTSP supplemented extenders have beneficial effects on at frozen-thawed drone sperm parameters. The results of the present study demonstrated the advantage of using a 10% RTSP supplemented extender for drone sperm motility, plasma membrane functionality, and mitochondrial function.Publication Sublethal effects of deltamethrin on apis mellifera in Turkey(Oxford Univ Press, 2015-04-01) Salazar, T. Antioxidant capacity, bioaccessibility of phenolics, hydrolysable and extractable phenolics, sugar, phenolic and fatty acid profiles were determined in the Spirulina honey. Total phenolic content of the Spirulina honey was 14.37 mg GAE/100g and increased 28.53% with Spirulina enrichment. Antioxidant capacities of the Spirulina honey were determined as 6.31, 5.85 and 3.66 mu mol Trolox/g by ABTS, CUPRAC and DPPH methods in bioaccessible extracts. No significant difference was observed between control and Spirulina honey samples in terms of bioaccessibility of the total phenolics (p <= 0.01). Among the detected phenolic compounds (15 phenolics), acacetin (48.55%) and pinocembrine (47.13%) were the highest. Total phenolic content of the honey can be enriched up to 20% by feeding of the honey bees with 10% of Spirulina. Saccharose content in Spirulina honey was lower than the control honey as 4.42%.Publication Drone semen cryopreservation with protein supplemented tl-hepes based extender(Kafkas Univ, Veteriner Fakultesi Dergisi, 2019-07-01) Alcay, Selim; ALÇAY, SELİM; Çakmak, Selvinar; Çakmak, İbrahim; ÇAKMAK, İBRAHİM; Mülkpınar, Emine; Toker, Mehmed Berk; TOKER, MEHMED BERK; Üstüner, Burcu; ÜSTÜNER, BURCU; Şen, Hasan; Nur, Zekariya; NUR, ZEKARİYA; Veteriner Fakültesi; 0000-0002-1438-221X; AAH-2635-2021; AAH-2558-2021; AAG-7238-2021; A-2794-2014The aim of the current study was to determine the optimum concentration of bovine serum albumin for post-thawing quality of drone sperm and this is the first study to evaluate the effect of BSA supplemented TL-Hepes based extenders for drone semen cryopreservation. Sexually mature drones were used for semen collection. Pooled semen was diluted with TL-Hepes based extender supplemented with different concentrations of BSA (1 mg/mL, 3 mg/mL, and 5 mg/mL) or without BSA (control), at a final concentration of 100x10(6) spermatozoon/mL. Motility, plasma membrane functional integrity (HOST), and defected acrosome (PSA-FITC) were evaluated in the study. At post thaw, the highest sperm motility rates were obtained in the BSA5 group (P< 0.05). Functional integrity of sperm membrane was better preserved in the BSA3 and BSA5 groups compared to the other groups. The acrosomal integrity rates were higher in BSA5 group than in control group (P< 0.05). The study shows that bovine serum albumin supplemented TL-Hepes based extenders have beneficial effect on drone semen parameters at post-thaw. The results of the study demonstrated a notable advantage of using 5 mg/mL of BSA in TL-Hepes based extender.Publication The investigation by PCR and culture methods of foulbrood diseases in honey bees in south marmara region(Kafkas Üniversitesi, 2015-01-01) Borum, A. In the present study, we investigated the chronic exposure of chemicals set (deltamethrin, acetamiprid, oxalic acid, fumagillin, and amitraz) on some sperm parameters and on the histomorphology of seminal vesicles. We also assessed the colony development and nosema load before and after the exposure. Thirty native Apis mellifera anatolica honeybee colonies with sister queens equalized with brood and total frame of bees were used for this experiment. Five colonies were used for each group. Deltamethrin, acetamiprid and fumagillin were given as oral chronic exposure at final concentrations of 25.10(-6) mg L-1, 0.01 m L-1 and 50 mg L-1 respectively in syrup solution (50/50). Colonies were exposed to oxalic acid by spraying 5 mL per frame space of 3% (w/v) of oxalic acid dihydrate. Finally, the amitraz was applied based on the manufacturer's instructions. The concentrations chosen represented the field-realistic concentrations and those used by beekeepers in the region. Results showed that deltamethrin reduced brood production. In the same group, we found a high increase in nosema load. All treatments decreased sperm count except for fumagillin but this compound increased sperm mortality and increased the percentage of sperm with defected acrosome integrity. The amitraz exhibited a high sperm mortality and high percentage of sperm with defected membrane integrity function. The sperm parameters such as the count, the motility, the acrosome integrity, the membrane function of sperm, and the histomorphology of seminal vesicles of drones exposed to oxalic acid were the most affected. Bee medications commonly used such as oxalic acid and fumagillin should be more investigated and should be considered by beekeepers and particularly queen breeders.Publication Antioxidant and bioaccessibility characteristics of functional fruit and vegetable honeys produced by innovative method(Elsevier, 2022-05-04) Demircan, H.; Tosunoğlu, H.; Çavus, Filiz; Şen, H.; Oral, Rasim Alper; Güldaş, Metin; GÜLDAŞ, METİN; Çakmak, İbrahim; ÇAKMAK, İBRAHİM; YILDIZ, ELİF; Gürbüz, Ozan; GÜRBÜZ, OZAN; Keleş Meslek Yüksekokulu; Gıda Mühendisliği Bölümü; 0000-0002-5187-9380; 0000-0003-3375-8777; 0000-0001-7530-7478; 0000-0003-1356-9012; 0000-0001-7871-1628; 0000-0003-1694-0104; U-1332-2019; AAK-4760-2020; GLT-7125-2022; AAG-6424-2021The functional fruit and vegetable honeys were produced by the natural bee feeding method with pomegranate, orange and black carrot concentrates. The natural enrichment method did not cause any negative effects on the quality criteria questioned in the standards such as moisture, pH, free acidity and HMF (Hydroxymethylfurfuraldehyde) content and the quality of the functional honeys obtained was high. Parallel to TPC contents, the antioxidant values of the honeys were increased 5.00-31.50, 16.09-53.96 and 21.18-39.72% by enrichment of orange, pomegranate, and black carrot concentrates. Gallic acid in pomegranate honey (14.52%) and chlorogenic acid in black carrot honey (27.01%) were determined as antioxidant components increased by enrichment. The enrichment process also led to an increase in the bioaccessibility characteristics. The increased antioxidant activity of functional honeys (DPPH) was verified by a high negative correlation value (R =-0.9103) and decreasing IC50 ratios in the opposite direction.
