Despite the exact mechanisms of these changes are not known, it is thought that ABT causes a decrease of CD4/CD8 ratio in the recipient. This study aimed to determine the CD4/CD8 ratio in stored erythrocyte suspensions (ES) and to obtain new information about TRIM mechanisms.Materials and Methods: Whole blood components used in our study were collected from 10 healthy volunteers. ES' obtained from whole blood were divided into three equal aliquots. Test samples which were related to 0th, 21st and 42nd storage days were prepared from these aliquots. CD3, CD4 and CD8 surface markers in peripheral blood mononuclear cells were investigated with flow-cytometer in these test samples. Results: Our data were evaluated according to storage days. Decrease of CD3 (p=0,001), CD4 (p<0,001), CD8 (p=0,012) expressing mononuclear blood cells and helper T cells in 21st-day samples, CD4 (p=0,035) and CD8 (p=0,017) expressing cells in 42nd-day samples compared to Day 0 samples and increase of CD3 (p=0,027) expressing cells in 42nd-day samples compared to 21st-day samples were found statistically significant. Conclusion: In our study, we did not find any significant change in the ratio of CD4(+)/CD8(+) cells in ES. Detailed studies can help us to obtain more comprehensive knowledge on this field.Publication Interleukin-21: A potential biomarker for diagnosis and predicting prognosis in covid-19 patients(Tubitak Scientific & Technological Research Council Turkey, 2021-01-01) ACET ÖZTÜRK, NİLÜFER AYLİN; URSAVAŞ, AHMET; GÖREK DİLEKTAŞLI, ASLI; DEMİRDÖĞEN, EZGİ; COŞKUN, NECMİYE FUNDA; EDİGER, DANE; UZASLAN, AYŞE ESRA; YÖYEN ERMİŞ, DİĞDEM; KARACA, MERT; TERZİ, ORKUN ERAY; BAYRAM, MERVE; ÖMER TOPÇU, DİLARA; Yigitliler, Busra; YURTTAŞ, AHMET; Maharramov, Shahriyar; YAZICI, GAMZE; ORAL, HALUK BARBAROS; KARADAĞ, MEHMET; Bursa Uludağ Üniversitesi/Tıp Fakültesi.; 0000-0002-6375-1472; 0000-0001-7099-9647; 0000-0002-7400-9089; 0000-0003-3604-8826; 0000-0002-2954-4293; 0000-0001-5871-8769; 0000-0003-0463-6818; 0000-0002-9027-1132; AAG-7406-2021; AAD-1271-2019; AAH-3888-2021; K-7285-2012; AAG-8744-2021; AAI-3169-2021; JPK-7012-2023; AAE-9142-2019Background/aim: COVID-19 patients have a wide spectrum of disease severity. Several biomarkers were evaluated as predictors for progression towards severe disease. IL-21 is a member of common gamma-chain cytokine family and creates some specific effects during programming and maintenance of antiviral immunity. We aimed to assess IL-21 as a biomarker for diagnosis and outcome prediction in patients hospitalized with COVID-19. Materials and methods: Patients with a preliminary diagnosis of COVID-19 and pneumonia other than COVID-19 admitted to a tertiary care hospital were included consecutively in this comparative study. Results: The study population consisted of 51 patients with COVID-19 and 11 patients with non-COVID-19 pneumonia. Serum IL-21 concentration was markedly higher, and serum CRP concentration was significantly lower in COVID-19 patients compared to nonCOVID-19 pneumonia patients. Within COVID-19 patients, 10 patients showed radiological and clinical progression. Patients with clinical worsening had lower lymphocyte count and haemoglobin. In addition to that, deteriorating patients had higher urea, LDH levels, and elevated concentration of both IL-6 and IL-21. The cut-off value of 106 ng/L for IL-21 has 80.0% sensitivity, %60.9 specificity for discriminating patients with clinical worsening. Multivariable analysis performed to define risk factors for disease progression identified IL-6 and IL-21 as independent predictors. Odds ratio for serum IL-6 concentrations >_ 3.2 pg/mL was 8.07 (95% CI: 1.3747.50, p = 0.04) and odds ratio for serum IL-21 concentrations >_ 106 ng/L was 6.24 (95% CI: 1.04 - 37.3, p = 0.02). Conclusion: We identified specific differences in serum IL-21 between COVID-19 and non-COVID-19 pneumonia patients. Serum IL-21 measurement has promising predictive value for disease progression in COVID-19 patients. These hypersensitivity reactions have been shown to be associated with the HLA-B*57:01 allele. High-resolution HLA-B*57:01 scanning has a time and cost disadvantage compared with low-resolution HLA-B*57 scanning. Before starting abacavir treatment, we will discuss whether high-resolution scanning is more beneficial in individuals who are positive on HLA-B*57 screening. This is the study with the largest cohort to investigate the prevalence of HLA-B*57 in Turkey. Methods The results of 25 thousand 318 people who applied to Bursa Uludag University Faculty of Medicine, Department of Immunology for HLA-B* typing were scanned. Results In our study, the HLA-B*57 serotype was detected in 827 (3.3%) individuals. Conclusion Considering these results, it can be assumed that the prevalence of HLA-B*57:01 in Turkey is lower than 3.3%. Instead of a high-resolution HLA-B*57:01 scan in all patients starting abacavir therapy, a high-resolution HLA-B*57:01 scan might be of greater benefit in patients who are positive on a low-resolution HLA-B*57 scan.Publication The use of ACE INDEL polymorphism as a biomarker of coronary artery Chock disease (CAD) in humans with Mediterranean-style diet(Elsevier, 2019-02-15) Temel, Şehime Gülsün; Ergören, Mahmut Çerkez; Yılmaz, İzel; Oral, Haluk Barbaros; TEMEL, ŞEHİME GÜLSÜN; Yılmaz, İzel; ORAL, HALUK BARBAROS; Bursa Uludağ Üniversitesi/Tıp Fakültesi/Tıbbi Genetik Anabilim Dalı.; Bursa Uludağ Üniversitesi/Sağlık Bilimleri Enstitüsü/Tıbbi İmmünoloji Anabilim Dalı.; Bursa Uludağ Üniversitesi/Tıp Fakültesi/İmmünoloji Anabilim Dalı.; 0000-0002-9802-0880; 0000-0002-1918-2516; AAG-8385-2021; JSK-9450-2023; K-7285-2012The ACE INDEL gene polymorphisms are strongly associated with CAD. Therefore, the present study was undertaken to investigate the relationship between ACE INDEL polymorphism and CAD in Turkish Cypriots whose are expected to have Mediterranean-style diet. 273 Turkish Cypriot descent volunteer subjects (186 controls and 87 CAD patients) participated in this study. Genotyping for the ACE INDEL polymorphism was performed by PCR-RFLP analysis. Biochemical parameters except the glucose and triglyceride lipid level were all within normal limits. Glucose level was found significant (p = 0.019) and triglyceride level was observed at the borderline for significance (p = 0.050) in participants according to WHO guidelines. With the respect to the genotype and allele distributions of ACE INDEL, the results showed statistically significant in CAD patients (p = 0.034) and not significant (p = 0.190) in controls. Haplotype analysis showed that D allele was more frequent in patients compared to controls. Thus, there is a statistically significant association with CAD disease with DD genotypes (p = 0.030) in Turkish Cypriot population. The results indicated that ACE INDEL polymorphism is an important predictor of coronary artery disease in Turkish Cypriots. Although 47% of the studied Turkish Cypriot population carry the D allele (p = 0.07), protocols should be developed for prevention strategies immediately. (C) 2018 Elsevier B.V. The vast majority of tumors are squamous cell carcinoma (SCC). Several genes encoding different cytokines may play crucial roles in host susceptibility to cancer because cytokine production capacity varies among individuals and depends on cytokine gene polymorphisms. Materials and Methods: The association between cytokine gene polymorphisms with primary laryngeal SCC was investigated. DNA samples were obtained from a Turkish population of eighty patients with primary cancer and fifty healthy controls. Results: All genotyping (interferon-gamma, transforming growth factor-beta 1 [TGF-beta 1], tumor necrosis factor-alpha [TNF-alpha], interleukin [IL]-6, and IL-10) experiments were performed using polymerase chain reaction sequence-specific primers. When compared to the healthy controls, the frequencies of TGF-beta 1 codon 25 (rs1800471) GC genotype and 25 C allele were significantly more common in the patient group. Conclusions: These results suggest that TGF-beta 1 gene polymorphisms may affect host susceptibility to laryngeal cancer.
