Item Bikomponent ve karışım hdpe / ldpe liflerinin eriyikten çekim yöntemi ile üretimi ve 3d yazıcılarda kullanımı(Bursa Uludağ Üniversitesi, 2019-08-26) Uslu, Oğuzhan; Aykut, Yakup; Bursa Uludağ Üniversitesi/Fen Bilimleri Enstitüsü/Tekstil Mühendisliği Anabilim Dalı.; 0000-0001-9132-0664; 0000-0002-5263-1985Termoplastik polimerlerin eritilerek yeniden şekillendirilebilmesi ve bünyesinde meydana gelebilecek deformasyonların ısıl işlemlerle giderilebilmesinden dolayı üç boyutlu (3D) yazıcılarda üretilip kullanımı son yıllarda yaygınlaşmıştır. Termoplastik polimerik yapılar önce eriyikten çekim yöntemi ile istenen özelliklerde lif formuna getirilip daha sonra bu liflerin 3D yazıcılarda kullanılması ile 3D yapılar elde edilmektedir. Bu kapsamda, termoplastik özellikte olan düşük yoğunluklu polietilen (LDPE) ve yüksek yoğunluklu polietilen (HDPE)’den farklı kontrüksiyonlarda saf, karışım ve bikomponent lifler eriyikten çekim yöntemi ile üretilmiştir. Üretilen liflerin kimyasal, kristal mikroyapı analizleri, termal stabiliteleri ve mekanik özellikleri incelenmiştir. Liflerin 3D yazıcılarda kullanılabilirliğini gözlemlemek için 3D yazıcıda numuneler üretilmiştir. Üretilen liflerden bikomponent yapıda olan LDPE / HDPE lifinin kullanılan 3D yazıcı için en uygun lif olduğu tespit edilmiştir. Bu liften 3D balpeteği yapı üretilip bu yapının bası mukavemeti PLA lifinden üretilen balpeteği yapı ile kıyaslanması suretiyle incelenmiştir. LDPE / HDPE bikomponent liften üretilen balpeteği yapının bası mukavemeti özellikleri piyasada yaygın kullanılan PLA’nın değerlerine yakın olduğu gözlemlenmiştir ve PLA gibi termoplastik karakterde olan bu lif yapısının PLA’ya alternatif olarak 3D yazıcılarda kullanılabileceği sonucuna varılmıştır.Item Catalytic graphitization and formation of macroporous-activated carbon nanofibers from salt-induced and H2S-treated polyacrylonitrile(Springer, 2013-05-23) Pourdeyhimi, Behnam; Khan, Saad A.; Aykut, Yakup; Uludağ Üniversitesi/Mühendislik Fakültesi/Tekstil Mühendisliği Bölümü.; 55320835000We present here a facile method to produce macroporous-activated carbon nanofibers (AMP-CNFs) by post-treating electrospun cobalt(II) chloride (CoCl2) containing polyacrylonitrile (PAN/CoCl2) nanofibers with hydrogen sulfide (H2S) followed by carbonization. A range of techniques including scanning and transmission electron microscopy, FTIR and Raman spectroscopy is used to examine and characterize the process. Because of the phase behavior between carbon and cobalt, cobalt particles are formed in the nanofibers, some of which leave the fibers during the heat treatment process leading to macroporous fibrous structures. The number of the macroporous increase significantly with increasing CoCl2 concentration in the precursor H2S-treated PAN/CoCl2 nanofibers. The cobalt phase in the fibers also leads to catalytic graphitization of the carbon nanofibers. Nanofibers morphologies transformed from uniform to bead on a string defect morphology with increasing Nafion ratio in CMA/N hybrid nanofibers. The melting point of CMA was detectable at DSC measurement, but since the addition of Nafion did not allow a proper crystallization of CMA, melting peak disappeared after the Nafion addition. Decomposition temperature decreased dramatically with the addition of Nafion into CMA/N nanofibers and decomposition took place at a broad temperature range. Nanofibers were also electrospun on the cylindrical graphite electrode for DNA electrochemical sensor analysis. Unmodified and NH-modified single strand DNA molecules were immobilized via physical adsorption method on the asprepared nanofiber sensory system. Electrochemical analysis were performed via differential pulse voltammetry (DPV) to observe the guanine oxidation signal at unmodified and NH-modified DNA. Maximum oxidation signals were detected from pure CMA nanofibers at unmodified DNA. Signal intensity increased with the addition of Nafion into CMA/N nanofibers at NH-modified DNA sample comparing to unmodified DNA. More enzymes can be immobilized on a surface when nanofibers are used as a supporting surface because the specific surface area increases tremendously. In this regard, polycaprolactam/cellulose monoacetate (PA6/CMA) and polycaprolactone/cellulose monoacetate (PCL/CMA) blended nanofibers (NFs) were prepared via an electrospinning process. Protease enzymes were immobilized on neat PA6, PCL, PA6/CMA, and PCL/CMA nanofibers and glutaraldehyde (GA) activated analogs through the physical adsorption method. The immobilized enzyme activity was measured by using a casein substrate, and the results were compared with free enzyme activity. Among all of the samples, the highest immobilization yield of about 82% was obtained with GA-activated neat PCL NF samples. The best remaining activity of the immobilized enzymes on pure CMA NFs was found to be 59% after seven reuses. Even after nine reuses, enzyme activities are still observed on the CMA NF samples. It was expected that the addition of CMA in PCL and PA6 NFs would increase the reusability number to reach the reusability of CMA NFs, but it was not significantly enhanced. Nanofiber (NF) diameters for pure CMA dramatically decreased from between 2.4 mu, -306 nm to between 958-42 nm with addition of hydrochloric acid (HCl) catalyzer into the electrospinning solution. Uniform CMA nanofibers morphologies transform to more defect structures containing particular structures with the addition and increase of TEOS content in CMA electrospinning solution. Also, nanofibers' diameters range became more fluctuated form and ultrafine nanofibers (diameters below 100 nm) existed more in the nanofiber mat. Even though the melting point was seen at CMA nanofibers, melting points were not detectable in CMA/TEOS NFs since TEOS addition does not allow an appropriate crystallization. Thermal analysis results revealed that residual contents after TGA measurement in nitrogen atmosphere were gradually increased by increasing TEOS ratio in CMA/TEOS nanofibers. ssDNA molecules were immobilized on the as-spun nanofibers and differential pulse voltammetry (DPV) measurements were carried out to investigate the guanine oxidation in ssDNA. Guanine oxidation signal intensities decreased with the initial addition of TEOS to CMA nanofibers and increases again by increasing TEOS content in CMA/TEOS hybrid nanofibers and then gradually decreases again with increasing TEOS content. Bu çalışmanın amacı nanoliflerin takviyesinin parafin aktivatörlerin ısıl histerezis özelliklerine etkisinin incelenmesidir. Öncelikle nanolif yüzeyi ve farklı karbon atom sayısına sahip sıvı parafinin etkileşimi elektroçekim yöntemiyle üretilmiş nanolif ile deneysel olarak değerlendirilmiştir. Tek iğneli uniaksiyel elektroçekim ile tek adımda hızlı ve kolay bir parafin/poliakrilonitril hibrid nanolif üretimi üzerine çalışmalar yapılmıştır. Üretilen hibrid parafin/poliakrilonitril yapısı parafin aktivatörün histerezis değerinin iyileştirilmesi için kullanılmıştır. Dört farklı erime noktasına sahip parafin çeşidi ile ağırlıkça %2.5, 5, 7.5 ve 10 oranlarında parafin/poliakrilonitril karışımı kullanılarak parafin aktivatör üretilmiş ve ısıl histerezis değerinde %15.3 oranına kadar iyileşme sağlanmıştır. Isıl histerezisde nanolif takviyesi ile elde edilen iyileşme, metal oksit nanoparçacık takviyesi ile kıyaslanmıştır. CuO, , ZnO, nanoparçacıkları ve poliakrilonitril nanolif takviyesi üretilen parafin aktivatör numuneleri, sırasıyla %24.6, 26.2, 20.0, 29.2 ve 30.8 oranlarında ısıl histerezis iyileştirmesi gözlenmiştir. Dispersiyon ısıl stabilitesinin, parafin aktivatörün histerezis performansına etkisini ölçmek için bu numuneler yüksek ısıda yaşlandırma testine tabi tutulmuştur. Silver NPs offer advantage over other materials for their effective antibacterial properties and being safer for humans at low concentrations. The synthesis of silver NPs may not always be environmental friendly and their incorporation in the polymer membranes is usually a multistep process. In this study, PVDF/PVP/AgNPs electrospun membranes were developed in a single step process where silver NPs were synthesized using reducing and stabilizing properties of PVP. The UV-vis spectroscopy confirmed the synthesis of silver NPs in PVP solution by sharp absorption peak at 398 nm. The membranes were loaded with various concentrations of silver NPs (1, 1.5, 2 and 2.5 wt%). The scanning electron microscopy of the developed membranes showed nano fibers of uniform diameter at optimized electrospinning conditions. FTIR spectroscopy also confirmed the successful development of polymeric composite (PVDF/PVP/AgNPs) membranes. The composite membranes demonstrated effective antibacterial properties against Staphylococcus aureus in disk diffusion test. The size of the inhibition halo increased with the concentration of the silver NPs in electrospun membranes. The findings of this study will be helpful in the simple and environmentally friendly development of antibacterial membranes for applications such as air and water filtration.Item Effects of surfactants on the microstructures of electrospun polyacrylonitrile nanofibers and their carbonized analogs(Wiley, 2013-12-05) Pourdeyhimi, Behnam; Khan, Saad A.; Aykut, Yakup; Uludağ Üniversitesi/Mühendislik Fakültesi/Tekstil Mühendisliği Bölümü.; 55320835000In this study, the influence of surfactants on the processability of electrospun polyacrylonitrile (PAN) nanofibers and their carbonized analogs was investigated. The surfactants employed in this effort are Triton X-100 (nonionic surfactant, SF-N), sodium dodecyl sulfate (SDS) (anionic surfactant, SF-A), and hexadecyltrimethylammonium bromide (HDTMAB) (cationic surfactant, SF-C). Interactions between electrospun PAN and the surfactants, reflected in effects on as-spun and carbonized nanofiber morphologies and microstructures, were explored. The results show that uniform nanofibers are obtained when cationic and anionic surfactants (surfactant free and nonionic surfactants) are utilized in the preparation of electrospun PAN. In contrast, a bead-on-a-string morphology results when the aniconic and cationic surfactants are present, and defect structure is enhanced with cationic surfactant addition. Moreover, fiber breakage is observed when the nonionic surfactant Triton X-100 is employed for electrospinning. After carbonizaition, the PAN polymers were observed to have less ordered structures with addition of any type of surfactant used for electrospinning and the disorder becomes more pronounced when the anionic surfactant is utilized. Owing to the fact that microstructure defects create midband gap states that enable more electrons to be emitted from the fiber, an enhancement of electron emission is observed for PAN electrospun in the presence of the anionic surfactant.Publication Electrospun mgo-loaded carbon nanofibers: Enhanced field electron emission from the fibers in vacuum(Pergamon-elsevier Science Ltd, 2013-02-01) Aykut, Yakup; AYKUT, YAKUP; Bursa Uludağ Üniversitesi/Mühendislik Fakültesi; JTV-2018-2023MgO-loaded electrospun carbon nanofibers (MgO/CNFs) were prepared by electrospinning a magnesium acetate containing polyacrylonitrile composite followed by stabilization under an air atmosphere at 280 degrees C and carbonization under a nitrogen atmosphere at 800 degrees C. In addition to investigating the morphological and material features of the nanofibers, the field emission (FE) characteristics of the carbonized NFs (CNFs), performed in an ultra-high vacuum chamber utilizing scanning electron microscopy (SEM), were determined. The results of the investigation show that the MgO/CNFs (195.5% enhancement) display enhanced field electron emission as compared to that of pure CNFs as a result of the existence of a MgO phase. Fibrous structures are some of the most commonly used materials for sound absorption applications. Enset fibers have a limited use in technical textile applications especially in packaging sector and are mostly left as an agro waste materials. The aim of this research work was to investigate the acoustic property of this new alternative natural fiber (fabric) and its green composite material. The sound absorption performance of enset fabric and its green composite was determined with impedance tube method. The porosity and void structure of enset fibers were studied by scanning electron microscope (SEM) for analysis of enset fabric and enset fabric reinforced composite sound dissipation mechanism. The sound absorption coefficient of enset fabric could only reach to 0.5 level with 5 layers of enset fabric after 3500 Hz frequency. Composite structures with increasing number of fabric layers decreased sound absorption frequency interval. With 5 layers composite (5 fabric layers in the composite), a sound absorption coefficient remained over 0.5 at a large frequency interval between 2830 and 6000 Hz while it was reaching to 0.9 at around 2500 Hz frequency. Increasing the ratio of bio resin to enset fabric caused the sound absorption behavior of composite material tend to shift from higher to medium frequency regions.Publication Green synthesis of cellulosic nanofiber in enset woven fabric structures via enzyme treatment and mechanical hammering(E.u. Enset is effectively un-utilized agro-waste fiber composed of cellulose, hemicellulose and lignin, which are resemblance to banana fibers. This paper focused on the utilization of enset waste materials for the production of nano fiber. Enset nano fiber (ENF) was fabricated by defibrillation of raw fibers from the surface of enset woven fabrics into micro and nano-scale fibers by a-amylase enzyme treatment and then mechanical hammering. Instead of sandwiching the nanofiber mats between conventional woven fabrics, in this research work, nanofiber mats were manufactured on the surface of the conventional woven fabrics. This top-down nanofabrication approach is simple, cost-effective and environmentally friendly nanofiber manufacturing technique from the surface of woven fabric structures. Fiber characterization was done by fourier transform infrared spectroscopy (FTIR) and scanning electron microscope (SEM). The surface area, pore size and pore volume of enset nano fiber was analyzed by brunauer, emmett and teller (BET). The test results revealed that nano fiber fabrication was significantly affected by the used enzyme concentration and applied mechanical power. Enset micro fiber was formed when 10 % (w/v) concentration of alpha-amylase enzymatic treatment was applied while enset nano fibers were formed when the enzyme concentration was increased to 15% - 20 % (w/v). The FTIR results revealed that hemicellulose and lignin were effectively removed and fine fibers were defibrillated from the crystal structure of enset woven fabrics. In this regard, two different approaches were performed. Aqueous sulfuric acid solutions were poured on the pure PAN nanofibers and heat treatment process was performed to determine the maximum temperature at which fiber structures does not deteriorate. Also, SA was directly added into electrospinning solutions with different ratios and heat treatment process was applied to the as-spun PAN/SA fibers. The fibers were produced both directly on a grounded plate and cylindrical pencil graphite surfaces (PGE) for the fiber characterizations and biosensor measurements. Single strand DNA (ssDNA) molecules were immobilized on the samples for the electrochemical investigation of guanine oxidation signal. 0.971 and 3.69 mu A guanine oxidation signal intensities were detected for neat PGE and heat-treated PAN/SA fiber coated PGE. The formation of sulfonic groups on the fibers has driven more ssDNA attachment on the surface, and hence increased the guanine oxidation signal intensity dramatically (about 261.4% increment). The biosensor was developed to detect Val66Met single point mutations in the brain-derived neurotrophic factor gene, which is frequently observed in neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, and bipolar disorder. The oxidation signal of the most electroactive and stable DNA base, i.e., guanine, was used at approximately + 1.0 V. Pencil graphite electrode (PGE) surfaces were coated with polyacrylonitrile nanofibers by electrospinning. Cyclic voltammetry was applied to the nanofiber-coated PGE to pretreat its surfaces. The application of cyclic voltammetry to the nanofiber-coated PGE surfaces before attaching the probe yielded a four fold increase in the oxidation signal for guanine compared with that using the untreated and uncoated PGE surface. The signal reductions were 70% for hybridization, 10% for non-complementary binding, and 14% for a single mismatch compared with the probe. The differences in full match, non-complementary, and mismatch binding indicated that the biosensor selectively detected the target, and that it was possible to determine hybridization in about 65 min. In the second route, PAN NFs were electrospun on a PGE surfaces and polymerization of PAni was conducted on the surfaces of the as-spun PAN NFs. NFs were kept at -18 degrees C in a refrigerator for several days. ssDNA was immobilized on the prepared NFs and guanine oxidation signals were observed for each system. The results revealed that use of PAN NFs enhanced signal intensity from 0.92 mu A (PGE) to 1.04 mu A (PAN NFs). Addition of PAni to PAN increased signal intensity to 1.23 mu A. When the PAN NF surfaces were coated with PAni, signal enhancement continued to increase up to 4.19 mu A for fourth day and decreased again when PAni-coated NFs were kept at -18 degrees C in the refrigerator. In this regard, hemoglobin assisted carbon nanofibers (CNFs) were prepared via electrospinning of the precursor polyacrylonitrile/hemoglobin (PAN/Hb) hybrid nanofibers and the following heat treatment process. Addition of low ratio Hb in the precursor PAN nanofibers caused a catalytic effect on the reaction taken place during the stabilization process that helps the formation of more graphitic structure during the carbonization process. But, increasing Hb ratio in the PAN/Hb nanofibers caused an inhibiting effect on the related reactions. Guanine oxidation signals of miRNA molecules were determined via differential pulse voltammetry (DPV) measurement. In this regard, the attachment of anti-miRNA molecules on the CNFs immobilized screen-printed electrodes (SPEs) and a following hybridization of the attached anti-miRNA with miRNA molecules were carried out. Three different miRNA molecules including the target (miRNA), single-base mismatched (SM.miRNA), and non-complementary (NC.miRNA) were hybridized with the previously attached anti-miRNA molecules on the Hb-CNFs immobilized SPEs. The enhancement of the guanine oxidation signal level was observed by using Hb-CNFs instead of using CNFs. That could be attributed to the increase of the graphitic level with low Hb addition to the precursor PAN/Hb nanofibers that causes a catalytic effect on carbonization process. They have focused on natural fibers as a reinforcing material having low cost, low density and good mechanical strengths. Enset woven fabric is manufactured from enset fibers, which are abundantly available, light weight (1.2 g/cm(3)), moderate moisture content (10%) and biodegradable un-utilized new fiber under Ensete Ventricosum family. This research work focused on investigating mechanical properties of a new green composite fabricated by enset woven fabric reinforcement and specially prepared bioresin from plant gums. The tensile strength of the enset fabric was improved four times from 528 to 2074 N in single-layer composite structure, while double-layer composite tensile strength was obtained as 3480 N. The test results indicated that the laboratory-made bioresin matrix from acacia and frankincense gums had a significant effect on the mechanical strength of composite structures compared with enset fabrics having the same number of threads (warps) in the specimens. Increasing the grammage of bioresin restored energy and improved the performance of tensile and flexural strength by reducing the deformation of composite structures. Generally, this study revealed that enset woven fabric as reinforcing structure and mixed acacia-frankincense bioresin as a matrix materials had a promising result to be used as composite materials in light weight structural applications with the same potential to improve its performance.Item Nafyon/selüloz asetat hibrit nanoliflerin üretimi ve elektrokimyasal biyosensörlerde kullanımının araştırılması(Bursa Uludağ Üniversitesi, 2019-07-01) Bostancı, Ayşe; Aykut, Yakup; Bursa Uludağ Üniversitesi/Fen Bilimleri Enstitüsü/Tekstil Mühendisliği Anabilim Dalı.Yapılan bu tez çalışmasında Selüloz monoasetat/Nafyon (CMA/N) hibrit nanolifler elektro çekim yöntemi ile üretilmiştir. Nanolifler, malzeme karakterizasyonları için düz alüminyum folyo üzerine, sade ve NH- ile modifiye edilmiş tek sarmal DNA (ssDNA)'nın elektrokimyasal guanin oksidasyon sinyal analizi için ise silindirik grafit elektrot üzerinde toplanmıştır. Maksimum Nafyon oranına sahip homojen nanolifler elde etmek için çeşitli oranlarda CMA/N nanolifler üretilmiştir. Nafyon oranının arttırılması homojen olmayan çözelti özelliğine yol açmış ve çözelti şeffaftan bulanık faza dönüşmüştür. Aynı zamanda nanoliflerin morfolojileri CMA/N hibrit nanoliflerde Nafyon oranının artmasıyla üniform yapıdan boncuklu lif haline dönüşmüştür. CMA'nın erime noktası, DSC ölçümünde saptanabilir, ancak Nafyon eklenmesi, CMA'nın uygun bir kristalizasyonuna izin vermemesi nedeniyle erime noktası Nafyon ilavesinden sonra tespit edilememiştir. CMA'nın termal bozunma sıcaklığı, CMA/N hibrit nanoliflere Nafyon eklenmesiyle önemli ölçüde azalmış ve bozunma geniş bir sıcaklık aralığında gerçekleşmiştir. Diferansiyel Darbe Voltametresi (DPV) analizi hazırlanmış olan sensör sistemi kullanılarak sade ve NH- modifiyeli DNA'da guanin oksidasyon sinyalini gözlemlemek için yapılmıştır. En yüksek oksidasyon sinyalleri saf ssDNA'da saf CMA nanoliflere immobilizasyonu sonrası elde edilmiştir. DNA'nın sinyal şiddetleri karşılaştırıldığında, NH ile modifiye edilmiş ssDNA numunesindeki CMA/N nanoliflere Nafyon eklenmesi ile sinyal yoğunluğunun NH ile modifiye edilmemiş ssDNA'lara kıyasla arttığı gözlemlenmiştir. Bunun sebebi ssDNA üzerindeki NH- grupları ile Nafyon'un yapısındaki SO3 gruplarının ve aynı zamanda CMA üzerindeki OH grupları ile ssDNA arasındaki hidrojen bağlarının bir etkileşiminde kaynaklanabileceği olarak yorumlanmıştır. Biyosensör yüzeyi ve ssDNA molekülü arasındaki güçlü etkileşim, sinyal artışına yol açmaktadır. İncelenen CMA/N hibrit nanolifler, DNA biyosensör cihazları için umut verici bir ara yüz olabilir.Item Nanolif kaplı kalem grafit biyosensör yüzeyinde spesifik nükleik asit dizilerinin hibridizasyonunun elektrokimyasal olarak tespiti(Uludağ Üniversitesi, 2016-06-28) Tanik, Nilay Aladağ; Demirkan, Elif; Aykut, Yakup; Uludağ Üniversitesi/Fen Bilimleri Enstitüsü/Biyoloji Anabilim Dalı.Bu çalışmada, nanolif kaplı grafit biyosensör yüzeyinde spesifik nükleik asit dizilerinin hibridizasyonunun elektrokimyasal olarak tespiti amaçlanmıştır. Bu amaçla, Alzheimer, Parkinson, Bipolar Bozukluk gibi nörodejeneratif hastalıklarda sıklıkla gözlenen nörotrofik faktörler ailesinden BDNF geninde meydana gelen Val66Met tek nokta mutasyonunun ilk kez elektrokimyasal tayinine yönelik bir biyosensör geliştirilmiştir. Bu polimorfizminin tayini için genelde kullanılmakta olan yöntemler, RFLP, RT-PCR ve DNA sekanslama yöntemleridir. Ancak bunlar pahalı yöntemler olup aynı zamanda oldukça zaman alan örnek hazırlama süreçleri gerektirmektedir. Tez kapsamında yapılan ön çalışmalarda, guanine dayalı yani indikatörsüz bir ayırımın mümkün olduğu görülmüş ve yöntem olarak DNA'daki en elektroaktif ve kararlı yanıt veren baz olan guanin bazının yaklaşık +1,0 V civarında verdiği yükseltgenme sinyaline dayalı indikatörsüz hibridizasyon algılama yöntemi kullanılmıştır. Literatürde ilk defa PGE yüzeyi elektrospin yöntemi ile nanolifle kaplanmıştır. Nanolif kaplı elektrot yüzeyine prob tutturmadan önce CV uygulanması, yüzeyden alınan guanin sinyalinin CV uygulanmamış ve nanolif kaplı olmayan PGE yüzeyine göre 4 kat arttışına sebep olmuştur. Çalışmada optimum koşulların belirlenmesinde tek sarmal DNA'da gözlenen guaninin oksidasyon sinyalinin, dsDNA(hibrit) ve tek bazı eşleşmemiş DNA(MM)'dan daha fazla olması esas alınmıştır ve mümkün olan en iyi ayırımın sağlandığı koşullar en iyi tayin koşulları olarak belirlenmiştir. Buna göre en uygun prob ve hedef tamponu 5X SSC, ölçüm tamponu ABS, prob konsantrasyonu 10µg/ml, hedef konsantrasyonu 15µg/ml, prob tutturma ve hibridizasyon süresi 30dk, hibridizasyondan sonraki yıkama süresinin 20sn olduğu görülmüştür. Tek sarmal DNA da yani hibridizasyon meydana gelmemiş dizilerde (prob, NC ve MM dizilerinde) guaninler açık olduğu için yükseltgenme sinyali yüksek, hibridizasyondan sonra ise sitozinle aralarında oluşturdukları hidrojen bağı köprüleri nedeniyle yükseltgenmesi kısmen kapalı duruma geldiğinden tek sarmala göre daha düşük sinyal vermesi beklenmiştir. Sonuç olarak tek sarmal olan prob diziden alınan sinyal en yüksek, hibrit diziden alınan sinyal en düşük, NC ve MM dizilerinden alınan sinyalin de hibridizasyon meydana gelmediği için prob dizisine yakın bir değerde olduğu görülmüştür. Ayrıca bu sonuç, geliştirilen biyosensörün seçimli şekilde hedefine bağlandığını ve polimorfizm tayinin mümkün olduğunu göstermiştir. Yöntemimizin kolay uygulanabilir olması, pahalı ekipmanlara gerek duyulmaması, hızlı cevap veren bir sistem olması (yaklaşık 65dk'da) ve herhangi bir toksik veya radyoaktif ajanın kullanılmaması nedenleriyle klasik yöntemlere güçlü bir alternatif yöntem geliştirilmiştir. In this regards, PVA/Cotton (C), PVA/Flax (F) and PVA/Wool (W) blended woven fabrics were prepared for the manufacturing fabric reinforced hydrogel composites. The hybrid woven fabric reinforced fabrics were treated with different concentrations of borax solutions. Aqueous borax solutions were used to alter the PVA yarns in the fabric into cross-linked structure in order to enhance mechanical performance of the hydrogel composite. Morphological investigation of hydrogel composites in a dried form was carried out by scanning electron microscopy (SEM) imaging. The chemical characterization of aqueous borax treated samples was examined by fourier-transform infrared spectroscopy(FTIR) measurements. Mechanical performances of the hydrogel composites were observed by tensile measurements. Thermogravimetric analysis (TGA) was conducted to characterize thermal stability of hydrogel composites. The results revealed that natural fiber woven fabric reinforcement significantly enhanced the mechanical strength of hydrogel composites, and wool fabric reinforced composite had better mechanical performance than its cotton and flax counterparts. Due to the low mechanical properties of hydrogels in general, the prepared fabric reinforced hydrogel composites could be used in hydrogel applications where mechanical strength is critically important.Item One-step preparation and investigation of high strength textile reinforced hydrogel composites(Bursa Uludağ Üniversitesi, 2020-12-25) Koç, Ümit; Aykut, Yakup; Eren, Recep; Bursa Uludağ Üniversitesi/Fen Bilimleri Enstitüsü/Tekstil Mühendisliği Anabilim Dalı.; 0000-0002-9061-3040Hidrojeller, ilaç salınımında, tarımsal alanda, biyomedikal alanda, gıda korumasında ve doku uygulamalarında etkin ve yaygın olarak kullanılan polimerik jellerdir. Hidrojellerin yumuşak ve esnek özellikte olması disiplinler arası yaklaşımın ilerlemesini önemli ölçüde sağlamaktadır. Fakat hidrojeller uygulanan mekanik kuvvetler altında düşük dirence sahiptir. Bu durum hidrojellerin kullanımı sırasında kırılmalara neden olmaktadır. Bu bağlamda, tekstil takviyeli hidrojel kompozitler hidrojellerin mekanik problemlerini çözmek için disiplinler arası bir yaklaşım sunmaktadır. Bu çalışmanın amacı mekanik özellikleri iyileştirilmiş dokuma kumaş takviyeli hidrojel kompozit yapıların incelenmesidir. Bu bağlamda, hidrojel iplik formunda olan polivinil alkol (PVA), uygulanan mekanik kuvvete karşı direncini artırmak için mukavemetli pamuk, keten, yün, viskon ipliklerle güçlendirildi. Bu nedenle, kumaş takviyeli hidrojel kompozitlerin üretimi için PVA/Pamuk, PVA/Keten, PVA/Yün, PVA/Viskon karışımlı dokuma kumaşlar hazırlandı. Hibrit dokuma kumaşlar, farklı oranlarda glutaraldehit ve boraks çözeltileri ile muamele edildi. Hibrit dokuma kumaşta bulunan PVA iplikleri çapraz bağlı jel yapılara dönüşürken, pamuk, keten, yün ve viskon iplikler, takviye elemanı olarak dokuma kumaşlarda kalmıştır. İplik ve kumaşla güçlendirilmiş hidrojel kompozitlerinin karakterizasyonları Fourier-transform kızılötesi spektroskopi (FT-IR), termogravimetrik analiz (TGA), X-ışını kırınımı (XRD), taramalı elektron mikroskobu (SEM) ile gerçekleştirildi. Hidrojel kompozitlerdeki çapraz bağlayıcı oranına ve su içeriklerine bağlı olarak numunelerin şişme dereceleri ve mekanik özellikleri incelendi. Tüm sonuçlar göz önüne alındığında, doğal lif takviyeli dokuma kumaşlar hidrojel kompozitlerin mekanik özelliklerini önemli ölçüde artırmıştır. Bu tezde sunulan yayımlanmış her makalenin bulguları, gelecekteki çalışmalar için birtakım önemli çıkarımlara sahiptir.
