dc.description.abstractBu çalışmanın amacı değişik sıcaklık dereceleri ve nem oranlarında (N.O.) Echinococcus granulosus protoskolekslerinin viabilite (canlılık) ve enfektivitelerinin incelenmesidir. Çalışmanın ilk kısmında, E. granulosus protoskolekslerinin viabiliteleri eosin vetrypan boyama yöntemleri ile incelenmiştir. En uzun canlı kalma süreleri;-10ºC ve % 50 N.O.'da 3 gün,0ºC ve % 60 N.O.'da 36 gün,+10ºC ve % 65 N.O.'da 28 gün,+20ºC ve % 70 N.O.'da 12 gün,+30ºC ve % 75 N.O.'da 4 gün,+40ºC ve % 80 N.O.'da 3 gün olarak belirlenmiştir. Araştırmanın ikinci bölümünde, ilk kısımda bildirilen sıcaklık dereceleri ve nemoranlarındaki protoskolekslerin enfektiviteleri deneysel enfeksiyon gerçekleştirmek suretiyle 7 grup köpekte incelenmiştir. Her sıcaklık ve nem grubu için, köpeklere değişik viabilite oranlarında yaklaşık 20.000 E. granulosus protoskoleksi sakatat kıyması içinde yedirilmiştir. Birinci grup -10ºC sıcaklık, % 50 N.O.'da % 5'i,kinci grup 0ºC sıcaklık, % 60 N.O.'da % 30'u,Üçüncü grup +10ºC sıcaklık, % 65 N.O.'da % 20'si,Dördüncü grup +20ºC sıcaklık, % 70 N.O.'da % 0'ı,Beşinci grup +30ºC sıcaklık, % 75 N.O.'da % 15'i,Altıncı grup +40ºC sıcaklık, %80 N.O.'da % 11'i,Kontrol grubu ise %68'i viabil olan protoskolekslerle enfekte edilmişlerdir. Enfeksiyondan sonraki 29-49'uncu günlerde nekropsileri yapılan birinci, dördüncü ve altıncı grup köpeklerde olgun E. granulosus'a rastlanmamış, ikinci gruptakilerden 200,250, 320; üçüncü gruptakilerden 25, 34, 39; beşinci gruptakilerden 25, 40, 56; kontrol grubundakilerden 1100, 2100, 1400 adet E. granulosus toplanmıştır.tr_TR
dc.description.abstractObserving the viability and infectivity of Echinococcus granulosus protoscolicesat different temparatures and humidities.The aim of this study was to examine the viability and infectivity of Echinococcusgranulosus protoscolices at different temparatures and humidities.In the first part of the study, viability of the E. granulosus protoscolices wereexamined by eosin and trypan staining. The maximum survival times were;3 days at -10ºC and 50 % Relative Humidity (R.H.),36 days at 0ºC and 60 % R.H.,28 days at +10ºC and 65 % R.H.,12 days at +20ºC and 70 % R.H.,4 days at +30ºC and 75 % R.H.,3 days at +40ºC and 80 % R.H..In the second part of the research, experimental infections were performed on the 7groups of dogs in order to examine the infectivity of protoscolices at varied temparaturesand humidities. Dogs were fed with offal mince harboring approximately 20.000protoscolices of E. granulosus at different viabilities. Dogs were infected with E.granulosus protoscolices of5 % viability at -10ºC and 50 % R.H. for the first group,30 % viability at 0ºC and 60 % R.H. for the second group,20 % viability at +10ºC and 65 % R.H. for the third group,0 % viability at +20ºC and 70 % R.H. for the fourth group,15 % viability at +30ºC and 75 % R.H. for the fifth group,11 % viability at +40ºC and 80 % R.H. for the sixth group,68 % viability for the control group.Zero E. granulosus at first , fourth and sixth groups; 200-250-320 E. granulosus atsecond group; 25-34-39 E. granulosus at third group; 25-40-56 E. granulosus at fifth groupand 1100-2100-1400 E. granulosus at control group were recovered from the necropsieddogs at 29-49 days after infection.en_US
