Abstract

This study was conducted to investigate comparatively various biochemical test on normal and subclinical mastitic milk samples, diagnosed according to the California Mastitis Test (CMT), Direct Microscopic Somatic Cell Count (DMSCC), Differential Cell Count (DCC) results, and to establish usefulness of these tests in the diagnosis of subclinical mastitis cases in cows. In the present study, a total of 100 Holstein, Brown-Swiss cows, in 3-7 years old, supplied by Faculty of Veterinary Medicine University of Uludağ and commercial dairy farms in the region of Bursa, were used as research materials. California Mastitis Test was performed previously on milk samples obtained from all cows. According to the test results, 30 mammary quarters shown (-) reactions were identified as free from subclinical mastitis. The other 70 mammary quarters reacted (?, +,++, +++) were identified as subclinical mastitis. The mean Somatic Cell Count (SCC) were 1.087.571 ±58.489 cells/ml in subclinical mastitic milk samples whereas 134.666±1 5.798 cells/ml in normal milk samples(P<0,001). Differential cell counts performed on normal milk samples indicated that epithelial cells 83,41 % of the total SCC while leucocytes 16,58 % of the total SCC. Subclinical mastitic milk samples 31,44 % epithelial cells and 68,55 % leucocytes. It was established that PMN leucocytes in normal and subclinical mastitic milk samples were 20,89 % and 81^20 % of total leucocyte counts, respectively. * The results of mineral elements analysis of milk serum samples collected from healthy and subclinical mastitis cows were shown as below; Na, 43,37±2,92 % mg'- 58,51^3,^8 % mg (P<0,005); K, 97,53±4,44 % mg - 80,37±4156 % mg (P<0,02); CI, 72,95±3,26 % mg - 108,27±4,50 % mg (P<0,001); Pi, 2,34±0,41 %mg - 2,29±0,04 % mg (P>0,90); Mg, 2;63±0,07 % mg - 2,51 ±0,04 % mg (P>0,20); Ca, 40f36±2,29 % mg - 49f05±1,85 % mg (P<0,05), respectively. The 3results of enzymes activity analysis of milk serum samples collected from healthy and subclinical mastitis cows were as below; LDH, 39,83110,98 U/L - 410,47±59,26 U/L (P<0;001); AST, 2,5±0,71 U/L - 22,74±4,92 U/L (P<0,02); ALT, 4,5±0,99 U/L - 12,23±1,88 U/L (P^OÖ); Amylase, 5/611,53 U/L - 6,67±1,86 U/L (P>0,50); GGT, 1326,38±160,37 U/L - 1710/71151,53 U/L (P>0,10); CK, 4,86±0,68 U/L - 14,04±1,91 U/L (P<0,005); ALP, 261,47±34,25 U/L - 723,79±64,66 U/L (P<0,001); ASP, 0,63±0,20 U/L - 2,16±0,34 U/L (P<0,005), respectively. The resuls of metabolytes analysis of milk serum samples collected healthy and subclinical mastitis cows were as below; glucose, 3,52±0,42 mg/dl - 1,53±0;15 mg/dl (P<0,001); cholesterol, 7,27±0,76 mg/dl - 8,04±0,57 mg/dl (P>0,20); triglyceride, 19;33±1,71 mg/dl -25,6411,81 mg/dl (P>0;05), respectively. As a result, it was concluded that direct and indirect somatic cell counts of milk, determination of the level of Na, K, Cl.glucose and LDH, AST, ALT, CK, ALP activities in milk serum samples can be used satisfactory! in the diagnosis of subclinical mastitis cases. Furthermore, if appropriate conditions are provided, AST enzyme activity of milk serum samples can be also used in diagnosis of subclinical mastitis cases.
