''New insights on neuronal nicotinic acetylcholine receptors as targets for pain and inflammation: A focus on α7 nAChRs''. Current Neuropharmacology, 16(4), 415-425.1570-159Xhttps://www.eurekaselect.com/article/85349https://hdl.handle.net/11452/40650Background: Nicotine and nicotinic acetylcholine receptors (nAChRs) have been explored for the past three decades as targets for pain control. The aim of this review is to introduce readers particularly to alpha 7 nAChRs in a perspective of pain and its modulation. Methods: Developments for alpha 7 nAChR modulators and recent animal studies related to pain are reviewed. Results: Accumulating evidences suggest that selective ligands for alpha 7 nAChRs hold promise in the treatment of chronic pain conditions as they lack many of side effects associated with other nicotinic receptor types. Conclusion: This review provides the reader recent insights on alpha 7 nAChRs from structure and function to the latest findings on the pharmacology and therapeutic targeting of these receptors for the treatment of pain and inflammation.eninfo:eu-repo/semantics/openAccessAlpha 7 nicotinic acetylcholine receptorsNicotinePainInflammationModulatorsAnti-inflammatoryAgonistsPositive allosteric modulatorCholinergic antiinflammatory pathwayGated ion-channelX-ray-structureNeuropathic painAnimal-modelsSpinal-cordBinding-sitesHuman monocytesDrug discoveryNeurosciences & neurologyPharmacology & pharmacyAnalgesicsAnimalsAnti-inflammatory agentsNon-steroidalCholinergic agentsHumansInflammationNeuronsPainReceptors, nicotinicNew insights on neuronal nicotinic acetylcholine receptors as targets for pain and inflammation: A focus on α7 nAChRsArticle0004319142000072-s2.0-8504894962641542516428820052https://doi.org/10.2174/1570159X15666170818102108NeurosciencesPharmacology & pharmacyInflammation; Methyllycaconitine; 3-(2,4-Dimethoxybenzylidene)AnabaseineBungarotoxin receptorGelatinase BGlial fibrillary acidic proteinHigh mobility group B1 proteinL kappa BImmunoglobulin enhancer binding proteinInterleukin 1Interleukin 10Interleukin 18Interleukin 1betaInterleukin 6Janus kinase 2Ligand gated ion channelMecamylamineMitogen activated protein kinase 1Mitogen activated protein kinase 3Myeloid differentiation factor 88Nicotinic receptorPhosphatidylcholineSerotoninSTAT3 proteinSuppressor of cytokine signaling 3Toll like receptor 4Tumor necrosis factorAnalgesic agentCholinergic receptor stimulating agentNicotinic receptorNonsteroid antiinflammatory agentAntiinflammatory activityBinding siteChronic painConformational transitionCytokine productionCytokine releaseDesensitizationDown regulationHumanInflammationMacrophage migrationNeuropathic painNonhumanPainPhosphorylationProtein expressionReviewSignal transductionUpregulationAnimalDrug effectInflammationMetabolismNerve cellPain1875-6190
