Autonomic Neuroscience: Basic and Clinical, 189, 50-55.1566-07021872-7484https://www.sciencedirect.com/science/article/pii/S1566070215000181https://hdl.handle.net/11452/40513The current study was designed to determine the effect of centrally administrated arachidonic acid (AA) on plasma gonadotropin hormone-releasing hormone (GnRH), follicle stimulating hormone (FSH), luteinizing hormone (LH) and testosterone level, and sperm parameters, and to show the mediation of the central cyclooxygenase (COX) to thromboxane A(2) (TXA(2)) signaling pathway in AA-induced hormonal and sperm parameter effects. Studies were performed in male Sprague-Dawley rats. A total of 150 or 300 mu l/5 mu l doses of AA were injected intracerebroventricularly (icv). AA significantly caused dose- and time-dependent increases in plasma FSH, LH and testosterone levels of animals, but not plasma GnRH level. AA also significantly increased sperm motility of the rats without change sperm number. Pretreated with ibuprofen, a nonselective COX inhibitor (250 mu g/5 mu l; icv), and furegrelate, a TXA(2) synthesis inhibitor (250 mu g/5 mu l; icv), prevented AA-evoked increase in plasma FSH, LH and testosterone levels, and sperm motility. In conclusion, our findings show that centrally administered AA increases plasma FSH, LH and testosterone levels and sperm motility of conscious male rats. Moreover, according to our findings, central COX-TXA(2) signaling pathway mediates these AA-induced effects.eninfo:eu-repo/semantics/closedAccessNeurosciences & neurologyArachidonic acidCyclooxygenaseThromboxane A(2)Hypothalamic-pituitary-gonadal axisSperm parametersIntracerebroventricularFollicle-stimulating-hormoneProtein-kinase-CLuteinizing-hormoneGonadotropin-secretionInsulin-secretionLipoxygenase productsProlactin-releaseHypotensive ratsBlood-pressureSperm qualityAnimalsArachidonic acidBenzofuransBrainDose-response relationship, drugEnzyme inhibitorsFollicle stimulating hormoneGrowth hormone-releasing hormoneIbuprofenInfusions, intraventricularLuteinizing hormoneMaleProstaglandin-endoperoxide synthasesRats, Sprague-DawleySignal transductionSperm countSperm motilityTestosteroneThromboxane A2Time factorsBrain thromboxane A2 via arachidonic acid cascade induces the hypothalamic-pituitary-gonadal axis activation in ratsArticle0003534274000082-s2.0-84925841026505518925784152https://doi.org/10.1016/j.autneu.2015.02.005Neurosciences & neurologyArachidonic acidFollitropinFuregrelateGonadorelinIbuprofenLuteinizing hormoneProstaglandin synthaseTestosteroneThromboxane A2Arachidonic acidBenzofuran derivativeEnzyme inhibitorFollitropinGrowth hormone releasing factorIbuprofenLuteinizing hormoneProstaglandin synthaseTestosteroneThromboxane A2AdultAnimal experimentArticleControlled studyFollitropin blood levelHormone blood levelHypothalamus hypophysis gonad systemLuteinizing hormone blood levelMaleNonhumanPriority journalRatSpermSpermatozoon motilityTestosterone blood levelAnimalBloodBrainDose responseDrug effectsIntracerebroventricular drug administrationMetabolismSignal transductionSpermatozoon countSprague Dawley ratTime
