Kalp yetmezlikli köpeklerde hemostazisin değerlendirilmesine yeni bir yaklaşım: Platelet proteomik profili

Abstract

Kalp yetmezliğinde plateletlerin aşırı artmış aktivasyonuna bağlı komplikasyonlar oluşabilmektedir. Hemostazis ve kardiyovasküler hastalıklar üzerine veteriner hekimlik alanında moleküler düzeyde yeterli çalışma bulunmamaktadır. Bu amaçla bu çalışmada doğal koşullarda kalp yetmezliği gelişen köpeklerde hemostazis (koagülasyon) değişimlerinin platelet proteomik profili ile detaylandırılması amaçlanmıştır. Bu çalışmada materyal olarak farklı ırk, yaş ve cinsiyetten toplam 20 köpek kullanılmıştır. Çalışmada kontrol grubu (n=10) ve test grubu (n=10) olarak iki grup oluşturulmuştur. Klinik, laboratuvar ve kardiyolojik muayene sonuçlarına göre sağlıklı olan köpekler kontrol grubuna, kronik kalp yetmezliği tanımlanan köpekler de test (hasta) grubuna dahil edilmiştir. Her bir köpekten platelet izolasyonu yapılmış ve pelletler analizlere kadar -80 derecede muhafaza edilmiştir. Platelet proteomları UPLC-ESI/QTOF/MS analizi ile belirlenmiştir. Platelet proteomları Canis lupus familialis'e göre tanımlanmıştır (n=107). Kontrole göre en az P<0,05 düzeyinde ve 1,2 kattan fazla değişim gösteren proteomlar (n=10) anlamlı kabul edilmişlerdir. Kontrole göre test grubunda guanine nucleotide-binding protein subunit alpha-11, apolipoprotein C-III, apolipoprotein A- II ve clusterin düzeylerinin arttığı; C-X-C motif chemokine 10, cytochrome C oxidase subunit 2, cathepsin D, serine/threonine-protein phosphatase PP1-gamma catalytic subunit, creatine kinase B-type ve myotrophin düzeylerinin ise azaldığı tespit edilmiştir. Sonuç olarak anlamlı değişim gösteren her bir platelet proteinin kalp yetmezliği sürecinde potansiyel biyobelirteç olabileceği ve bu nedenle bu proteinlerin saha çalışmaları ile validasyona ihtiyaç duyduğu kanısına varılmıştır.
Heart failure can cause haemostatic complications due to platelet over- activation. There is not enough information on the molecular relationship between hemostasis and cardiovascular diseases in veterinary medicine. Thus, in this study, it was aimed to elaborate hemostasis (coagulation) changes with the platelet proteomic profile in dogs with naturally occurred heart failure. The material of this study consisted of totally 20 dogs with different breed, age, and sexes. Two different group were designed; control (n=10) and test groups (n=10). Based on the physical, laboratory and cardiologic examination results, dogs were included into control (healthy) group, or dogs with heart failure were enrolled into test group. Platelet isolations were performed from each dog, and platelet pellets were stored -80 C until analysed. Platelet proteomes were identified by use of UPLC- ESI/QTOF/MS method. Platelet proteomes were matched with the previously described proteins for Canis lupus familialis, and compared to the control, at least P <0.05 level and 1.2 fold change of proteomes was considered statistically significant. Compared to control group, guanine nucleotide-binding protein subunit alpha-11, apolipoprotein C-III, apolipoprotein A-II and clusterin levels increased whereas CXC-motif chemokine-10, cytochrome-C-oxidase subunit-2, cathepsin-D, serine/threonine- protein phosphatase PP1-gamma catalytic subunit, creatine kinase B-type and myotrophin levels decreased in test group. As a result, each platelet protein identified in the present study could be a potential biomarker in the process of heart failure and therefore these proteins need validation with field studies.
