Abstract
Antiinsülin ve insülin bağlanmasıyla meydana gelen antikor-antijen kompleksinden radyoimmünolojik deneylerle faydalanılarak kanda insülin seviyesi tayini metotlarında serbest ve bağlı insülin fraksiyonlarının separasyonu son derece önemli bir konudur. Bu separasyon için zaman alıcı, kompleks ve masraflı metotlar ileri sürülmüştür. Bizim bu çalışmamızda oldukça kolay yapılabilen ve ufak bir kağıt şeritle yükselen kromatografi tekniği uygulayarak bahis konusu separasyonun mümkün olduğu gösterilmiştir. Ayrıca bu metotla altı kişide ağız ve damar içi yollarıyla yapılan glükoz yükleme testleri esnasında kanda insülin seviyesinin, ağız yoluyla verilenlerde daha belirgin tarzda yükseldiği de saptanmıştır.
Separation of free from antibody bound hormon is a major step in the radioimmunoassay method for the measurement of serum insulin level. Most of the methods require elaborate and expensive apparatus, and are very time - consuming. In this paper a simple modification of the chromatographic immuno-assay of insulin is described, using small,dry, vertical chromatographic paper wicks placed directly in the reaction mixture. Insulin secretion during the glucose tolerance is estimated on six different persons (seven sera during oral glucose tolerance test and seven sera during intravenous glucose tolerance test in every cases). The results are showing more insulin secretian in oral tests than intravenous ones. The rise in blood insulin levels following i.v. glucose is smaller than that found after oral administration.
