Publication Bazı balık türlerinden ve kum midyelerinden (Venus gallina) Vibrio parahaemolyticus i̇zolasyonu ve i̇dentifikasyonu(TÜBİTAK, 2002) Aydın, A.; Soyutemiz, Ece; Veteriner Fakültesi; Besin Hijyeni ve Teknolojisi Ana Bilim Dalı; 6504329566This study was performed to establish techniques used in the isolation and identification of Vibrio parahaemolyticus and investigate the presence of V. parahaemolyticus in fish and clams. Between June 1998 and June 2000, a total of 46 fish samples including 14 anchovies, 12 whiting, 10 horse mackerel and 10 sardines collected from fish markets in Bursa and a total of 70 clam samples collected from shellfish stations in Sakarya and Kocaeli districts were tested for V. parahaemolyticus. Fish samples were found to be free of V. parahaemolyticus. It was detected that total isolation from clams was 10 (14.3%). Methods and Results: Salmonella detection limits of the optimized cPCR were determined with DNA templates from the samples of tetrathionate broth (TTB), Rappaport Vassiliadis broth (RVB) and selenite cystine broth (SCB) artificially contaminated with 10-fold dilutions of 6 x 10(8) CFU ml(-1) of pure Salmonella enterica ssp. enterica serovar Enteritidis 64K stock culture. Detection limits of cPCR from TTB, RVB and SCB were found as 6, 6 x 10(1) and 6 x 10(4) CFU ml(-1), respectively. In addition, detection limits of bacteriology were also determined as 6 CFU ml(-1) with TTB and SCB, and 6 x 10(1) CFU ml(-1) with RVB. A total of 200 samples, consisting of 100 chicken and 100 turkey meat samples, were tested with optimized cPCR and bacteriology. Eight and six per cent of the chicken meat samples were found to harbour Salmonella by cPCR and standard bacteriology, respectively. Of six Salmonella isolates, four belonged to serogroup D, two to serogroup B. Conclusions: The TTB cultures of both artificially and naturally contaminated samples were found to be superior to those of RVB and SCB cultures in their cPCR results. This cPCR, utilizing template from 18-h TTB primary enrichment broth culture, takes approximately 40 min in the successful detection of Salmonella from poultry meat. Significance and Impact of the study: This study shows that cPCR from TTB enrichment culture of poultry meat would enable rapid detection of Salmonella in laboratories with low sample throughput and limited budget.Publication Sığır mastitislerinden izole edilen Escherichia coli suşlarında genişlemiş spektrumlu beta-laktamaz aktivitesi ve antibiyotik dirençlilik profilinin incelenmesi(Ankara Üniversitesi, 2012) Dinç, Gökçen; Ata, Zafer; Temelli, Seran; Uludaǧ Üniversitesi; Veteriner Fakültesi; Besin Hijyeni ve Teknolojisi Ana Bilim Dalı; W-7994-2019; AAI-1092-2021; 42360917900; 6506404118Bu çalışmada Ankara, Balıkesir ve Çorum’daki süt işletmelerinden sağlanan mastitis şüpheli sütlerden izole edilen 92 adet E. coli suşunda genişlemiş spektrumlu beta-laktamaz (GSBL) aktivitesi ve antibiyotik direnç profilinin araştırılması amaçlandı. E. coli suşlarında Kirby Bauer disk diffüzyon yöntemi ile GSBL tarama testi, fenotipik GSBL doğrulama testi ve 12 adet antibiyotik için in vitro duyarlılık testleri yapıldı. İncelenen E. coli suşlarında en yüksek dirençlilik oranları sırasıyla eritromisine (%69.6), ampisiline (%39.1), tetrasikline (%34.8), nalidiksik aside (%25.0), kloramfenikole (%22.8), trimetoprim-sülfametaksazole (%21.7) ve amoksisilin klavulonik aside (%21.7) karşı bulunurken, suşların %25.0’inin kullanılan tüm antibiyotiklere duyarlı olduğu saptandı. Ayrıca E. coli suşlarının %54.3’ünün iki veya daha fazla sayıda antibiyotiğe dirençli olduğu belirlendi. Bu çalışma ile ülkemizde mastitis orijinli E. coli suşlarında ilk kez GSBL aktivitesi araştırıldı. Ancak, incelenen E. coli suşlarında GSBL aktivitesi saptanmadı. Çalışmada elde edilen bulgular mastitise neden olan E. coli suşlarında yüksek oranda çoğul ilaç direncinin geliştiğini gösterdi.
