Item Comparison of serum IgG concentration, total protein, glutaraldehyde coagulation test and gamma glutamyl transferase in neonatal foals(Bursa Uludağ Üniversitesi, 2018-04-15) Kasap, Sevim; Kennerman, Engin; Babaeski, Serdar; Dulger, Hüseyin; Uludağ Üniversitesi/Veteriner Fakültesi.; 0000-0001-9501-9983; 0000-0003-3877-2893The purpose of the present study was to detect the passive transfer status in healthy neonatal foals by comparing serum immunoglobulin G (sIgG) concentration, serum total protein (STP), glutaraldehyde coagulation test (GCT) duration and gamma glutamyl transferase (GGT) activity . Fifteen neonatal foals (0-15 days old)’ blood samples were collected before suckling (day 0), 12th hour (hr), 24th hr (1st day), 7th and 15th days. Serum IgG and STP levels significantly increased after the 12th hr. Conversely, serum GCT duration significantly decreased (p< 0.05) in neonatal foals after the 12th hr. The result of the present study was shown that measurements of sIgG, STP concentration and GCT duration are useful parameters to detect Failure of Passive Transfer (FPT) in neonatal foals. The aim of this study was to determine the effects of a monensin controlled-release capsule administered 3 weeks before calving on blood metabolites (serum beta-hydroxybutiric acid (BHBA), cholesterol (CHOL), urea, creatinine (CREA), gamma glutamyl transferase (GGT), aspartate aminotransferase (AST), total protein (TP), phosphorus (P), magnesium (Mg+), calcium (Ca++), glucose (GLU) and plasma non-esterified fatty acids (NEFA) concentration) and milk yield before and after calving. A total of 50 Holstein-Friesian cows were selected from the same flock. Blood samples were taken 3 weeks before the expected calving date and during weeks 1, 2, 4, 6, and 8 after calving. The cows were divided into two groups: a study group (Group M, n = 40) and a control group (Group C, n = 10). Group M received orally a cylindrical device (Kexxtone, ELANCO Animal Health, Guelph, ON, Canada), and group C received no treatment. BHBA and NEFA were lower (P < 0.05) in group M compared with group C in weeks 6 and 8 after calving. GLU was higher (P < 0.05) in group M compared with group C in week 4 after calving. Urea was higher (P < 0.05) in group M compared with group C in week 1 after calving. There was no statistically significant difference (P <0.05) between the two groups in milk yields. Although milk yield was similar in both groups, the changes in the concentrations of BHBA, NEFA, GLU and urea show that a monensin CRC could be used as a glycogenic precursor.Publication Evaluation of glutaraldehyde coagulation test and colostrum brix refractometer compared with snap foal igg test in neonatal foals(Wiley, 2023-10-01) Babaeski, Serdar; Kasap, Sevim; KENNERMAN, ENGİN; KASAP, SEVİM; Kennerman, Engin; ORMAN, ABDÜLKADİR; Orman, Abdulkadir; TEMİZEL, ETHEM MUTLU; Yıldırım, Kübra Nur; YILDIRIM, KÜBRA NUR; Bursa Uludağ Üniversitesi/Veteriner Fakültesi; 0000-0001-9138-4422; AAG-9134-2021Background: Prompt diagnosis of passive transfer failure in the neonatal period is important for early treatment.Objectives: To compare the diagnostic performance of serum glutaraldehyde coagulation test (GCT) and colostrum BRIX% for failure to transfer passive immunity (FTPI) diagnosis with the results of SNAP foal test and to evaluate the results of serum GCT and colostrum BRIX% measurements in foals with diarrhoea in the 0-1 month period.Study design: In vitro experiments.Methods: Excess serum and colostrum (n: 298) from samples collected from newborn foals and their dams for clinical purposes were used. Foals were classified as FTPI positive (IgG < 8 g/L) or negative (IgG >= 8 g/L) using the SNAP foal test. We compared the sensitivity and specificity of serum GCT and colostrum BRIX % for diagnosing FTPI in all foals and in the sub-group of foals which developed diarrhoea within the first month of life was noted. The relationships between the results of the serum GCT and colostrum BRIX% and diarrhoea in foals with and without FTPI were evaluated.Results: Serum GCT and colostrum BRIX % were statistically significantly different (p < 0.05) between the foals without FTPI and with FTPI classified according to the SNAP test. Using a cut-off value for serum GCT of >10, sensitivity was 100% (95% CI 92.9%-100%) and specificity 100% (98.3%-100%) while with a cut-off value of <= 24, with colostrum BRIX% of <= 24 sensitivity was 92% (80.9%-97.8%), and specificity was 98% (95.3-99.3). In the sub-group of foals without FTPI using a colostrum BRIX% cut-off value of <= 26 the sensitivity for prediction of diarrhoea in the 0-1 month period was only 72.4% (52.8-87.3, p < 0.001) with specificity 54.3% (47.6-61.1) but the test performance was not robust (ROC AUC 0.61).Main limitations: The number of repeated measurements in the evaluation of serum GCT, and colostrum BRIX% was low. More clinical problems could be examined.Conclusions: The serum GCT, and colostrum BRIX%, both economical and practical to use in the field, gave results comparable with the SNAP foal IgG test. The ability to accurately predict diarrhoea in the first month of life with these tests was limited.Item Taylarda sütten kesilme sürecinde immunmodülatör uygulamasının etkilerinin değerlendirilmesi(Bursa Uludağ Üniversitesi, 2023-02-24) Babaeski, Serdar; Kennerman, Engin; Bursa Uludağ Üniversitesi/Sağlık Bilimleri Enstitüsü/Veteriner Fakültesi/İç Hastalıkları Anabilim Dalı.; 0000-0003-3877-2893Bu çalışmada başlıca stres kaynağı olduğu bilinen taylarda sütten kesme döneminde, immunmodülatörden Parapoxvirus ovis (PPVO) uygulaması yapılarak stres sonucu oluşabilecek klinik yansımalarının azaltılması veya önlenebilmesi hedeflendi. Bunun yanında bu süreçte oluşan stres sonrasında gelişebilecek yangısal reaksiyonları belirlemek için akut faz proteinlerinden olan serum amiloid A (SAA), serum kortizol, hücresel bağışıklık sistemindeki değişimleri izlemek adına sitokinlerden interferon gama (IFN-γ) ve interlöykin-10 (IL-10) düzeylerindeki değişimlerin değerlendirilerek elde edilen bu sonuçların klinik yansılmalarla ilişkilendirilmesi ve saha koşullarına uygunluğunun araştırılması amaçlandı. Çalışma grubu (n=20) için belirlenen ayrılma gününden 2 gün önce, ayrılma günü (0. gün) ve ayrılma sonrası 7. gün inaktif Parapoxvirus ovis içeren ticari preparattan (Zylexis®, Zoetis) tay başına 2 ml kas içi uygulandı. Kontrol grubuna (n=10) ise aynı günlerde serum fizyolojik enjekte edildi. Kan örnekleri -2, 0, 1, 7 ve 14. günlerde alındı. SAA konsantrasyonları -2., 0. ve 7. günlerde gruplar arası anlamlı olarak farklı tespit edilirken, IFN-γ kontrol grubunda bütün analiz günlerinde anlamlı olarak çalışma grubuna göre yüksek olduğu kaydedildi. Serum kortizol düzeyleri kontrol grubunda, çalışma grubuna kıyasla 1., 7. ve 14 günlerde yüksek olarak tespit edildi. Sonuç olarak, taylarda sütten kesme sürecinde özellikle IFN-γ ve kortizol düzeylerindeki değişikliklerin oluşan stresin belirlenmesinde önemli iki biyobelirteç olduğu saptandı. Sütten kesme döneminde immunmodülatör uygulaması yapılan grupta günlük canlı ağırlık artışında olumlu yansımaların olduğu, vücut sıcaklığının daha kontrollü seyrettiği ve taylar üzerinde oluşan stresin etkilerinin azaldığı kaydedildi. Tayların annelerinden ayrılacağı dönemde immunmodülatör uygulamasının taylarda stres sonucu oluşabilecek ekonomik kayıpların önlenmesinde etkili olduğu ve at yetiştiriciliğinde tayları sütten kesme protokolünde yer almasının uygun olacağı kanısına varıldı.
