Abstract

S. cerevisiae CYC1 geni sitokrom-c, isoform 1 proteinini kodlamaktadır. CYC1 geni insandaki sitokrom C1 geni ile yüksek oranda benzerlik göstermektedir. CYC1 aynı zamanda iso-1-cytochrome c olarak da bilinmektedir, mitokondride membranlar arası boşlukta bulunur ve mitokondriyel elektron taşıma zincirinin önemli bir bileşenidir. İnsan hücrelerinde sitokrom C1 proteinin mitokondriyel hasarlara yanıt olarak apoptozda yer aldığı bilinmektedir. İnsan CYC1 geninin komplementasyon ile S. cerevisiae'da işlevsel olduğu gösterilmiştir. Bazı stres şartlarına yanıt olarak apoptozun mayalarda da meydana geldiği daha önce hem hücresel ve hem de moleküler seviyede gösterilmiştir. S. cerevisiae Cyc1 proteininin bu maya türünde apoptoz süresicindeki işlevleri kesin olarak ortaya konulamamıştır. Bu araştırmada S. cerevisiae'da apoptoza neden olan bazı stres koşullarının CYC1 geni transkripsiyonuna ve S. cerevisiae hücre morfolojilerine olan etkileri incelendi. CYC1 geni glukoz baskılaması ile kontrol edildiğinden CYC1 geni ekspresyonu hem repres ve hem de derepres şartlarda incelendi. Oksidatif stresin CYC1 transkripsiyonunu yaklaşık % 25 kadar baskıladığı bulundu. S. cerevisiae'da apoptozu uyaran en önemli madde olan Salisilik asitin ise CYC1 transkripsiyonunu tamamen durdurduğu görüldü. Nitrosatif stresin ise CYC1 transkripsiyonunda baskılamaya yol açtığı belirlendi. Salisilik asitin genel olarak hücre morfolojilerinde değişime yol açtığı tayin edildi. Apoptoz uyarıcı maddelerin üreme ortamına uygulanması ile S. cerevisia hücrelerinde parçalanma başta olmak üzere, hücre morfolojisinde değişimler, hücre döngüsünün durması gibi anormallikler belirlendi.
CYC1 gene of S. cerevisiae encodes cytochrome-c, isoform-1 protein. CYC1 gene shows very high homology to human cytochrome C1 gene. CYC1 also known as iso-1-cytochrome c. Cyc1 protein is located at the intermembranal space of mitochondria and has a significant function in the electron transport chain. Human Cyt-C1 protein has a significant function in apoptosis that occurs in response to mitochondrial damages. It is known that human CYC1 gene complements CYC1 function in S. cerevisiae. Apoptosis also occurs in yeasts in response to various stress conditions. Molecular components of apoptosis signaling pathway has been characterized in S. cerevisiea. Nonetheless, the molecular functions of Cyc1 protein in yeast apoptosis has not been characterized in details yet. In this study, the effects of certain stress inducing agents on the transcription of CYC1 and cell morphology were investigated. Since the transcription of CYC1 is regulated by glucose repression, its transcriptional regulation was analyzed both in glucose repressed and derepressed growth conditions. It was found that the both nitrosative and also oxidative stress repressed CYC1 transcript by 25% under glucose derepressed conditions. Salicylic acid, which triggres apoptosis in yeast, completely inhibited CYC1 transcription. In addition, our results indicated that presence of apoptosis inducing chemicals, such as salicylic acid, in the growth medium resulted in significant changes in cell morphology and cell cycle arrest.
