Abstract

Son yarım asırda gıdalarda küflerin önemi giderek artmıştır. Bu önem, bir yandan küflerin ve özellikle küf metabolitlerinin gıda sanayinde artan kullanımı; diğer yandan küflerin bozucu etkileri yanında, aşırı toksik etkili ikincil metabolitler olan mikotoksinleri salgılamalarından kaynaklanmaktadır. Bu nedenle gıda ve gıda hammaddelerinin küflenmeye karşı korunmaları büyük özen gerektirmektedir. Bu çalışma, gıda koruyucusu olarak sıklıkla adı geçen propiyonik asit ve kalsiyum propiyonatın, gıda kaynaklı küfler üzerine etki derecelerini ve gerçekten engelleyici etkilerinin olup olmadığını belirlemek amacıyla planlanmıştır. Araştırma materyali olarak kullanılan küf materyalleri U. Ü. Ziraat Fakültesi Gıda Mühendisliği Bölümünde yapılan çalışmalarda, değişik gıda maddelerinden izole edilen ve tanılanan küf kültürleri arasından seçilmiştir. Bu amaçla seçilen türler Aspergillus niger, A. oryzea, A. flavus, A. nidulans, A. versicolor, Penicillium citrinum, P. verrucosum, P. echinulatum, P. roquefortii, P. islandicum, Fusarium semitectum, Mucor racemosus, M. circinelloides, Rhizopus oligosporus ve Cladosporium macrocarpum'dur. Besiyeri olarak PDA (Patates-Glikoz-Agar) ve CA (Czapek Agar) kullanılmıştır. Ca- propiyonat PDA'da 1, 1.5, 2, 2.5, 3 g/L konsantrasyonlarında ve pH 4.8, 5.8, 6.8'de denenmiştir. Propiyonik asit CA besiyerinin pH'sını 5.8 ve 4.8'e düşürecek şekilde, 0.1 ve 0.5 g/L düzeylerinde kullanılmıştır. Deneme sıcaklığı ise 28 °C olarak seçilmiştir. Propiyonik asit 0.1 g/L konsantrasyonunda, gelişme üzerine fazla bir engelleyici etki gösterememiş, hatta A. nidulans, A.-11- oryzea, A. versicolor, P. verrucosum, P. citrinum, P. echinulatum, P. islandicum, M. racemosus ve M. circinelloides türlerine gelişmenin erken veya geç dönemlerinde teşvik edici etki yapmıştır. 0.5 g/L propiyonik asit yalnızca Fu sari um semitectum türüne baştan sona kadar etkin bir durdurucu etki göstermiştir. Ca- propiyonat A. niger'e en fazla engelleyici etkiyi 6.8 pH'da %67 oranında gerçekleştirmiştir. Diğer türlere en fazla etki pH 4.8'da belirlenmişse de, bu hiç bir zaman A. niger türünde belirlenen seviye ye ulaşamamıştır. Durdurucu etki 1 g/L 'den2 g/L'ye kadar belirgin bir şekilde artmış, 2 g/L 'de3g/L'ye kadar ya artış olmamış veya önemsiz sayılabilecek düzeyde kalmıştır. Sonuç olarak, doğrudan gıda bileşimine katılacak düzeydeki propiyonik asit veya kalsiyum propiyonatın, tek başına gıdaları küf gelişimine karşı koruyucu olarak kullanılmalarının güvenli olmadığı saptanmıştır.
The importance of fungi has been increasing for a half the last century. There are several reasons concerning this development such as, increasing use of fungi and their metabolites and secondary toxical metabolites of fungi called mycotoxins. Therefore special care will be taken in the protection of food and food raw materials against spoilage caused by fungi. This study was conducted to determine the inhibitory effects and inhibition efficiency of propionic acid and calcium propionate on the food related fungi. The strains used in this study were selected from the strains which are isolated and identified in the examination of different food samples in the Food Engineering Agricultural Faculty, Department of Uludağ University. The selected species are: Aspergillus niger, A. oryzea, A. flavus, A. nidulans, A. versicolor, Penicillium citrinum, P. verrucosum, P. echinulatum, P. roquefortii, P. islandicum, Fusarium semitectum, Mucor racemosus, M. circinelloides, Rhizopus oligosporus ve Clodosporium macrocarpum. PDA( Potato- Glucose Agar) and CA(Czapek Agar) were used as growth media. Ca-propionate was used in PDA at different concentrations: 1, 1.5, 2, 2.5, 3g/ 1 with different pH values: 4.8, 5.8, 6.8. To be able to decrease the pH of CA growth medium to 5.8 and 4.8, PA concentrations were adjusted 01 g/ I and 0.5g/ I respectively. The Incubation temperature was chosen as 28 °C. Propionic acid with concentration 0.1g/ I has shown a low inhibitory effect on the growth. Propionic acid has shown even a positive effect on the growth of A. nidulans, A. oryzea, A. versicolor, P. verrucosum, P. citrinum, P. echinulatum, P.-IV- izlandicum, M. racemosus ve M. clrcinelloides both in the early or late incubation period. Propionic acid with 0.5 g/ I has shown an efficient inhibitory effect only on the growth of Fusarium semitectum during the whole study. During the study, maximum inhibitory effect was observed with pH 4.8. Whereas for A.nlger, the maximum inhibitory effect (%67) was observed with Ca- propionate with a pH 6.8. In the study, it was found that for inhibition at pH 4.8 was always less efficient than inhibition at pH 6.8 for A.nlger. Inhibitory effect was enhanced reasonably with an increase in concentration of Ca-propionate from 1g/ I to 2g/ I. The effect was negligible in the increase of concentration of Ca- propionate from 2g/ 1 to 3 g/ 1. Ultimately, it has been found that the use of Propionic acid or Ca- propionate alone is not safe for the preservation of foods against spoilage caused by fungi.
