Abstract

Bu çalışmada, tavuk ve hindi etlerinden Salmonella aranması amacıyla kapiller polimeraz zincir reaksiyonu (kPCR) geliştirildi. Çalışmanın ilk basamağında, saf Salmonella enterica subsp. enterica serovar Enteritidis 64K kültürü ile yapay kontamine Tetrathionate Broth (TTB), Selenite Cystine Broth (SCB) ve Rappaport Vassiliadis (RVB)birincil zenginleştirmelerinden alınan örneklerden hazırlanan 'templeyt'lerle optimize edilen kPCR'unun Salmonella arama limitleri saptandı. kPCR'unun arama sınırı TTB,RVB ve SCB ile sırasıyla 6, 60 ve 6x104 cfu ml-1 bulundu. Aynı şekilde, BAM(Bacteriological Analytical Manual)'da tanımlanan standart bakteriyolojinin saptama sınırı TTB ve SCB ile 6 cfu ml-1 ve RVB ile 6x10 cfu ml-1 olarak belirlendi. Optimize edilen kPCR ve standart bakteriyoloji ile 100 tavuk, 100 hindi eti örneği olmak üzere toplam 200 örnek test edildi. kPCR ve standart bakteriyoloji ile tavuk etlerinden sırasıyla, % 8,bakteriyoloji ile %6 oranında Salmonella saptandı. Tavuk ve hindi etlerinin zenginleştirme kültürlerinden PCR için templeyt hazırlamada, TTB'un SCB ve RVB'a üstünlüğü gözlendi. zole edilen 6 Salmonella suşunun 4'ü Serogrup D, 6'sı Serogrup B'ye aitti.Çalışmanın sonunda, TTB ile birincil zenginleştirme kültürlerinden templeyt hazırlanarak uygulanan ve toplam 25 dk zaman alan bir kapiller PCR optimize edildi ve kanatlı etlerinden Salmonella aranması amacıyla başarıyla uygulandı.
In this study, capillary PCR (cPCR) was developed for Salmonella detection fromchicken and turkey meat. At the first step of this study, Salmonella detection limits of theoptimized cPCR were determined with the DNA templates from the samples of Tetrathionate Broth (TTB), Selenite Cystine Broth (SCB), Rappaport Vassiliadis Broth(RVB) artificially contaminated with the pure Salmonella enterica subsp. enterica serovar Enteritidis 64K culture. Detection limits of cPCR with TTB, RVB, SCB were found to be6, 60 and, 6x104 cfu ml-1, respectively. In addition, detection limit of standard bacteriology described in BAM (Bacteriological Analytical Manual) was also determined as 6 cfu ml-1with TTB and SCB, and 6x10 cfu ml-1 with RVB. A total of 200 samples, consisting of100 chicken and 100 turkey meat samples were tested with the optimized cPCR and thestandart bacteriology. Salmonella was detected from the chicken meat samples at the ratesof 8 %, and 6 % by the cPCR and the standart bacteriology, respectively. Superiority ofTTB over SCB and RVB was observed in the PCR template preparation from enrichmentcultures of chicken and turkey meat samples. Of the isolated 6 Salmonella strains, 4belonged to serogroup D, 2 to serogroup B. Finally, cPCR using the template from TTBprimary enrichment broth culture and taking total 25 min was developed and this PCR testwas successfully used for Salmonella detection from poultry meat samples in this study.
