Abstract

Bu çalışmada sumakın (Rhus coriaria L.) farklı (aseton, dietil eter, etil alkol, etil asetat, kloroform, metanol ve su) ekstraktlarının antibakteriyel ve antifungal etkileri incelenmiştir. Bu amaçla disk difüzyon ve tüp seyreltme yöntemleri kullanılmıştır. Denemede yer alan mikroorganizmalar için, Minimum İnhibitör Konsantrasyon (MİK) ve Minimum Bakterisidal Konsantrasyon (MBK) veya Minimum Fungisidal Konsantrasyon (MFK) değerleri belirlenmiştir. Test bakterileri olarak Bacillus cereus, Enterococcus faecalis, Escherichia coli ATCC 25922, E. coli ATCC 35218, E. coli O157, E. coli TipI, Listeria monocytogenes, Proteus mirabilis, Salmonella enteritidis ATCC 13076, Salmonella enteritidis, Staphylococcus aureus ATCC 33862, Yersinia enterocolitica kullanılmıştır. Bunlar arasından, Y. enterocolitica ve S. aureus ATCC 33862 sumak ekstraktlarına en duyarlı türler olarak belirlenmişken, en dirençli türlerin Bacillus cereus ve Escherichia coli ATCC 25922 olduğu saptanmıştır. Bakteriler üzerindeki MİK ve MBK aralığının sırasıyla 5-150 ve 8-600 mg/mL olduğu kaydedilmiştir. Antibakteriyel etki bakımından en güçlü ekstraktın metanollü örnek olduğu tespit edilmiştir. Denemede antifungal aktivite 12 maya (Candida albicans ATCC 10231, C. oleophila, Kloeckera apiculata, Kluyveromyces lactis, K. marxianus, Metschnikowia fructicola, Pichia angusta, P. anomalo, Saccharomyces cerevisiae1, S. cerevisiae2, S.uvarum ve Schizosaccharomyces pombe) ve 13 küf (Alternaria alternata, Fusarium semitectum, F. oxysporium, Aspergillus fumigatus, A.niger ATCC 16604, A. niger, A. parasiticus1, A. parasiticus2, A. oryzae, A. versicolor, Penicillum chrysogenum, P.citrinum ve P. roqueforti) türü üzerinde test edilmiştir. Sumak ekstraktlarına en duyarlı ve dirençli mayalar sırasıyla P. anomalo ve C. albicans ATCC 10231 olarak kaydedilmiştir. İnhibisyon etkisi bakımından aseton ekstraktının mayalar üzerinde en güçlü etkiyi oluşturduğu, buna karşılık en zayıf aktivitenin kloroform ve sulu ekstraktlar tarafından açığa çıktığı saptanmıştır. Mayalar üzerinde belirlenmiş MİK ve MFK düzeyleri sırasıyla 4–2200 ve 6–2800 mg/mL olarak kaydedilmiştir. Bununla birlikte, genellikle sumak ekstraktlarının küfler üzerinde zayıf antifungal etki gösterdiği belirlenmiştir. Test küfleri arasından A. niger ATCC 16604 en dayanıklı tür olarak belirlenmişken, A. alternata’nın ise en duyarlı olduğu tespit edilmiştir. En düşük MİK ve MFK değerleri 250 ve 400 mg/mL olarak belirlenmişken, en yüksek dozda (3000 mg/mL) bile engellenemeyen türlerin olduğu saptanmıştır. Küfler üzerinde engelleyici etkiye sahip çözücü olarak, metanolün en başarılı ekstraktı verdiği gözlenmiştir.
In this study, different extracts (acetone, diethyl ether, ethyl alcohol, ethyl acetate, methanol, chloroform and water) of sumac (Rhus coriaria L.) were used to determine the antibacterial and antifungal effect. It was used disk diffusion and macrodilution method. Minimum inhibitory concentration (MIC) and Minimum Bactericidal Concentration (MBC) or Minimum Fungicidal Concentration (MFC) of all samples were determined. As test bacteria, Bacillus cereus, Enterococcus faecalis, Escherichia coli ATCC 25922, E. coli ATCC 35218, E. coli O157, E. coli Type I, Listeria monocytogenes, Proteus mirabilis, Salmonella enteritidis ATCC 13076, Salmonella enteritidis, Staphylococcus aureus ATCC 33862, Yersinia enterocolitica were used. Among these bacteria S. aureus ATCC 33862 and Y. enterocolitica were the most resistant strains whereas B. cereus and E. coli ATCC 25922 were the most sensitive. MIC and MBC values of sumac extract were 5-150 and 8-600 mg/mL, respectively. The highest antibacterial activity was exhibited by methanol extract. Antifungal activity was determined against 12 yeast- Candida albicans ATCC 10231, C. oleophila, Kloeckera apiculata, Kluyveromyces lactis, K. marxianus, Metschnikowia fructicola, Pichia angusta, P. anomalo, Saccharomyces cerevisiae1, S. cerevisiae2, S.uvarum ve Schizosaccharomyces pombe- and 13 fungi- Alternaria alternata, Fusarium semitectum, F. oxysporium, Aspergillus fumigatus, A.niger ATCC 16604, A. niger, A. parasiticus1, A. parasiticus2, A. oryzae, A. versicolor, Penicillium chrysogenum, P.citrinum ve P. roqueforti- species. Among the yeasts, P. anomalo ve C. albicans ATCC 10231 were determined as the most sensitive and resistant microorganisms, respectively. Acetone extract indicated the best inhibition effect, whereas chloroform and aqueous extracts of sumac had low activity against test yeasts. MIC and MFC levels were recorded 4–2200 and 6–2800 mg/mL, respectively. In addition, generally sumac extracts showed weak antifungal effect against fungi. In present study, A. niger ATCC 16604 was the most resistant strain to all test extracts while A. alternata was susceptible. The lowest MIC and MFC values of test fungi were 250 and 400 mg/mL. On the other hand, it was observed that maximum dose (3000 mg/mL) also had no inhibition effect some fungi. Among extracts which have inhibition effect, methanol was determined the best one against fungi.
