Abstract

Bu çalışmada sekiz blastomerli fare embriyolarında aspirasyon biyopsisi ve sekiz hücre aşamasından in vitro gelişen blastosistlerde trofektoderm biyopsisi yapıldıktan sonra, manipüle edilen embriyoların in vitro gelişim oranları, kalite değerlendirmesi ve alıcı farelere transferden sonra in vivo implantasyon ve fetal gelişim oranları araştırıldı.CB6F1 (C57BL ?6 X BALB?C) dişi farelere 10 IU gebe kısrak serum gonadotropini (SIGMA- PMSG) intraperitoneal (i.p) enjeksiyonla uygulandı. Enjeksiyondan 48 saat sonra, 7,5 IU insan koriyonik gonadotropini (Organon-hCG) i.p. yolla verilerek süperovulasyon protokolü tamamlandı ve dişi fareler erkek CB6F1 (C57BL ?6 X BALB?C) fareler ile çiftleştirildi. Süperovule dişiler hCG uygulamasından 68-72 saat sonra, sakrifiye edildi. Sakrifiye edilen farelerin oviduktları HEPES tamponlu ve 3 mg/ml BSA ile takviye edilmiş HTF medyumu ile yıkanarak 8 hücreli embriyolar elde edildi.Biyopsi işlemleri içerisinde Ca2+/Mg2+ bulunmayan HEPES tamponlu ve 3 mg/ml BSA + 5 µg/ml sitohalazin B içeren Quinn's HTF medyumunun 50 µl'lik damlalarında yapıldı. Blastomer ve trofektoderm biyopsi gruplarındaki embriyolar sırasıyla 48 saat ve 24 saat süre ile 4 mg/ml BSA içeren Quinn's blastosist medyumunda %5 CO2, %5 O2 ve 37°C yüksek nemli inkübatör içerisinde blastosist aşamasına kadar kültür edildi ve stereo mikroskop altında in vitro gelişim oranları değerlendirildi. Gelişen blastosistlerden bazılarında toplam hücre sayısı belirlenirken, bazılarıda alıcı CD-1 farelere transfer edildi ve 13-15 gün sonra in vivo gelişim oranları saptandı. Sonuçlar SPSS 17.0 istatistik programında bağımsız T-Testi ve ANOVA ile değerlendirildi.Blastomer aspirasyon grubunda 152 ve kontrol grubunda 63 sekiz hücreli embriyo kullanıldı ve in vivo gelişimi değerlendirmek için blastomer grubundan 36 ve kontrol grubundan 30 embriyo transferi yapıldı. Trofektoderm grubunda 79 ve kontrol grubunda 28 blastosist kullanıldı ve in vitro kültür sonrası in vivo gelişimi değerlendirmek için, trofektoderm grubundan 32 ve kontrol grubundan 22 embriyo transferi yapıldı. Gelişen blastosistlerden bazıları floresan boyama tekniği ile boyandıktan sonra, toplam hücre sayıları belirlendi.Blastomer biyopsi grubunda gelişim oranı %81,02 (121/152) ve toplam hücre sayısı 50 olarak saptanırken, kontrol grubunda ise gelişim oranı %96,37 (62/63) ve ortalama toplam hücre sayısı 50 olarak saptandı. Blastomer biyopsi ve kontrol gruplarının gelişim oranları arasında anlamlı bir fark bulunmadı. Gelişen blastosistlerin toplam hücre sayıları karşılaştırıldığında da gruplar arasında benzer biçimde anlamlı bir farka rastlanmadı. Blastomer biyopsi grubunda uygulanan embriyo transferi sonucunda %25 (9/36) oranında implantasyon bölgesi saptandı ve bu bölgelerde %19,44 (7/36) oranında fetal gelişim saptandı. Kontrol grubunda yapılan embriyo transferi sonucunda ise %26 (8/30) oranında implantasyon saptandı ve bu bölgelerde % 20 (6/30) oranında fetal gelişim belirlendi. İn vivo gelişimi değerlendirmek için yapılan embriyo transferi sonuçları karşılaştırıldığında, implantasyon alanları ve fetal gelişim oranları bakımından gruplar arasında istatistiksel bir fark saptanmadı.Trofektoderm biyopsi grubunda gelişim oranı %86,96 (69/79) ve toplam hücre sayısı 26,66 olarak saptanırken, kontrol grubunda gelişim oranı %93,33 (23/28) ve toplam hücre sayısı 55,33 olarak saptandı. Trofektoderm biyopsi ve kontrol gruplarının gelişim oranları arasında anlamlı bir fark saptanamamıştır. Gelişen blastosistlerin toplam hücre sayıları karşılaştırıldığında gruplar arasında anlamlı bir fark bulunmuştur (P<0,05). Trofektorderm biyopsi grubunda yapılan embriyo transferi sonucunda %21,88 (7/32) oranında implantasyon bölgesi saptandı fakat bu bölgelerde fetal gelişim oluşumu gözlenmedi. Kontrol grubunda yapılan embriyo transferi sonucunda ise %59,09 (13/22) oranında implantasyon bölgesi saptandı ve bu bölgelerden % 18,18 (4/22) oranında fetal gelişim saptandı. İn vivo gelişimi değerlendirmek için yapılan embriyo transferi sonuçları karşılaştırıldığında, implantasyon alanları bakımından gruplar arasında fark bulunmazken, fetal gelişim oranları bakımından gruplar arasında anlamlı bir fark bulundu (P<0,05).Çalışma sonucunda trofektoderm biyopsi grubunda in vitro gelişim ve in vivo implantasyon oluşumunun olumsuz etkilenmemesine karşın, in vivo fötal gelişim olumsuz yönde etkilenmiştir. Bunun olası nedeninin biyopsi sürecinde oluşan fazla sayıdaki hücre kaybı düşünülmektedir. Oysa, blastomer biyopsi grubunda fetal gelişim de dahil kontrol grubu ile aralarında anlamlı fark bulunmadı. Dolayısıyla, erken gelişim döneminde yapılacak biyopsi işlemlerinin embriyo gelişimini olumsuz yönde etkilemediği ve özellikle hızlı gelişen ve toplam hücre sayısı daha düşük olan fare gibi türlerde yapılacak biyopsi uygulamalarının erken gelişim dönemlerinde yapılmasının daha avantajlı olacağı sonucuna varıldı.
In this study, in vitro development ratios, quality evaluation, in vivo implantation and fetal development ratios after transfer to foster mother mice of manipulated embryos were investigated following aspiration biopsy in eight blastomer mouse embryos and trophectoderm biopsy in blastocyst developed from eight cell stage embryos in vitro.10 IU pregnant mare?s serum gonadotrophin (PMSG) was intraperitoneally injected to female CB6F1 (C57BL ?6 X BALB?C) mice and 48 hours later 7,5 IU human chorionic gonodotrophin (hCG) was also injected intraperitoneally to complete superovulation protocol. They were mated with male CB6F1 (C57BL /6 X BALB/C) mice. Superovulated female mice were approximately sacrificed 68-72 hours after hCG administration. Eight cell embryos were flushed from oviducts of sacrificed mouse with HTF (human tubal fluid) supplemented with HEPES and 3 mg/ml BSA.All embryos were biopsied in 50 µl drops of Ca2+/Mg2+ free HTF medium containing HEPES + 3 mg/ml BSA + 5 µg/ml cytochalasine B. After biopsy, embryos were cultured for 24 and 48 hours in Quins blastocyst medium supplemented with 4 mg/ml BSA in %5 CO2, % 5 O2, and 37°C in humidified air for blastomere biopsy and trophectoderm biopsy groups, respectively. After culture period, in vitro development ratios of embryos were evaluated under stereo microscope. Some of the developing blastocysts were used to determine total cell number and some of them were transferred to the recipient CD-1 mouse to determine in vivo development ratios at the 13-15th day of pregnancy. Results were evaluated by independent T Test and ANOVA of SPSS 17.0 statistic program.In blastomere aspiration and control groups, 152 and 63 eight cell embryos were respectively used. To evaluate in vivo development, 36 and 30 embryos at blastocyst stage were transferred from blastomere biopsy and control groups, respectively. In trophectoderm and control groups, 79 and 28 eight cell embryos were respectively used. To evaluate in vivo development, 32 and 22 embryos at blastocyst stage were transferred from trophectoderm biopsy and control groups, respectively. Some of developing blastocysts were stained with fluorescent staining technique and total cell numbers were determined.In blastomere biopsy and control groups, development rates and total cell numbers were determined as 81.02% (121/152), and 96.37% (62/63) and 50, and 50, respectively. There was not any significant difference between groups in terms of development ratios and total cell numbers. In blastomere biopsy and control groups, implantation region and fetal development rates were found as 25% (9/36), and 26% (8/309 and 19.44% (7/36), and 20% (6/30), respectively. No significant difference was observed between groups in terms of implantation region and fetal development rates.In trophectoderm biopsy and control groups, development rates and total cell numbers were found as 86.96% (69/79), and 93.33% (23/28) and 26.66, and 55.33, respectively. Although there was not any significant difference between groups in terms development rates, there was a significant difference between groups in terms of total cell numbers (P<0.05). In trophectoderm biopsy and control groups, implantation region and fetal development rates were determined as 21.88% (7/32), and 59.09% (13/22) and 0% (0/32), and 18.18% (4/22), respectively. Although there was not any significant difference between groups in terms implantation region rates, there was a significant difference between groups in terms of fetal development rates (P<0.05).As a result of this study, in trophectoderm biopsy group, while in vitro development and in vivo implantation formation were not affected negatively, in vivo fetal development is negatively affected. As a possible reason for this, it is thought that lots of cell losses during biopsy happen. However, there was no significant difference between groups in blastomere biopsy and control groups including fetal development rates. Therefore, it was concluded that biopsy applied at early stage of embryonic development does not affect embryo development negatively and biopsy procedures applied at early developmental stages have more advantages especially in embryos developing faster with low number of total cell number such as mouse species.
